Objective: To study the effects of pentabrominated diphenyl ethers on the structure and function of reproductive system in rats and mechanism.Methods: In time-effect study of pentabrominated diphenyl ethers on Male Reproductive System of Rats, 40 health 4 weeks old SD rats were divided into 8 groups randomly. Each group has 5 rats. These groups infected 5,15,30,45 and 40 days in the experimental and control groups respectively; Treated rats were gavaged daily with 120mg/kg BDE-99 in corn oil according to 0.4% body weight. Serum E2, T were measured in duplicate by standard radioimmunoassay kits, the pathology changes of thyroid tissue were observed by hematoxylin-eosin(HE) dyeing, and PCNA expression was examined by immunity histochemistry. In dose-effect study, 40 health 4 weeks old SD rats were divided into 4 groups randomly;The treated rats were orally administered various doses of BDE-99(60,120,180mg/kg), we examined the change of serum hormone, histopathology, histochemistry and follicular cells ultramicroscopic structure after 30 days.Results: Male SD rats Time-effect study showed: there were no difference between the experimental group and the control group at 5 days; at 15 days, testis organ coefficient of experimental group increased, E2 decreased, strength reduction in the expression of PCNA, Compared with the control group, those indicators are significant, small quantities of sperm within the twisted tangle; The experimental animals were exposed to 30 days of testis organ coefficient significantly higher in the experimental group, T, E2 concentrations decreased, PCNA expression was further reduced, some primary spermatocytes cells expressed a degeneration seminiferous epithelium, add sperm twisted tangles; at 45 days, the experimental group decreased organ coefficient, T, E2 concentrations lower, the difference is significant. Histopathologic examination revealed serious degeneration of seminiferous epithelium, sperm tangles serious. PCNA expression is further reduced by only part of spermatogonial cells. Dose-effect study showed: With increasing doses, testis organ coefficient increased, serum hormone levels lower, PCNA, ER, Arom, Bcl-2 expression reduced, Bax expression increased, activity of SDH, LDHc4 decreased, and the degeneration of seminiferous epithelium. These indicators are significant dose-effect relationshipResearch of female SD rats show: with increasing doses, serum T increased, E2 decreased; ovarian weight reduced; the expression of PCNA, ER, Bcl-2 reduced; expression of Bax increased; Luteal hyperplasia; the number of follicular were significantly reduced; Electron microscopy detect showed, Ovarian Granulosa Cell Apoptosis; Chromatin condensation, margination ; The follicle mesenchyme gland increased, The mesenchyme cell enlarges the memorial polygon, Common mitochondria of cytoplasm inner is surrounded by the grease.Conclusion: DE-99 can lead to damage of male reproductive system. It can reduce the serum sex hormone level; lead to reproduced secreting function of testis. It also can lead to female reproductive system loss. That whose mechanism may all be that DE-99 has restrained Arom activity, has used the conversion obstacle between the T and E2, affected testis and ovary's growth , The reproduction and secretion function using testis and the ovary have accepted effect.
