| Objective: To study the han Chinese and uighurs in urumqi cytochrome oxidase in neonatal umbilical cord blood P4501A1(CYP1A1) gene and tiny heterologous dimers partners(SHP) gene polymorphism and polybrominated diphenyl ethers(PBDEs)exposure levels and low birth weight(LBW).Methods:Case-control study method,selection and urumqi in xinjiang autonomous region people’s hospital maternity and child health care of 200 cases of maternal and neonatal epidemiological investigation by questionnaire, collecting neonatal umbilical cord blood and extract the DNA, and adopts wafergen high-throughput capture detection platform, the second generation sequencing technology SHP and CYP1A1 gene polymorphism;using isotope dilution and high resolution gas chromatography/high resolution double focusing magnetic type mass spectrometry(HRGC/HRMS) method to detect pregnant women venous serum 7 kinds of polybrominated diphenyl ether monomer concentration(BDE- 28, BDE- 47, BDE- 99,BDE- 100, BDE- 153, BDE- 154, BDE- 183), analysis and evaluation of exposure levels;using ELISA to detect pregnane receptor(PXR) levels in neonatal umbilical cord blood.Results:(1) Single factor analysis showed that only the SHP gene polymorphism is associated with the han low birth weight, the difference was statistically significant(P <0.05), multi-factor analysis showed that two kinds of genetic polymorphism has nothing to do with the han, d low birth weight(P > 0.05).(2) In the maternal venous blood Σ PBDEs content is 144.67 to 50037.29pg/g lw, contributing to PBDEs main source BDE- 153. Han Chinese mother in serum of different weight newborn BDE- 47, BDE- 100, Σ PBDEs content difference had statistical significance(P < 0.05); Uighur mother in serum of different weight newborn BDE- 47, BDE- 153, Σ PBDEs content difference hadstatistical significance(P < 0.05).(3) Multiple factors analysis shows that control the related influencing factors, when control the traditional influence factors of low birth weight, other factors unchanged BDE- 47, BDE- 100, the concentration of BDE- 154 with the han nationality in the pathogenesis low birth weight(P < 0.05);BDE- 100, BDE-153 concentrations are associated with Uighur low birth weight(P < 0.05).(4) the Uighur low birth weight PXR levels higher than normal birth weight(P < 0.05); The han normal weight and low birth weight PXR levels were higher than Uighur(P < 0.05).Conclusion:(1) CYP1A1 and SHP gene polymorphism have different effect on the occurrence of low birth weight(2) Han Chinese and uighurs were detected in maternal serum PBDEs, and able to han Chinese and uighurs PBDEs different monomers of low birth weight newborn caused a certain degree of influence.(3) the pregnane receptor(PXR) levels in maternal serum low birth weight has nothing to do with the han nationality, related to the Uighur low birth weight.(4) the occurrence of low weight is caused by the joint action of multiple factors, so in the future study, should as far as possible into a more comprehensive factors comprehensive analysis, in order to fully understand the causes of low birth weight occur,so as to improve the quality of newborn babies born. |
