Backgroud: Human tumor is an cell cycling disease. As an important positive regulator of cell cycle and chemis-drug resistant regulatory factor of tumor cell, Y box-binding protein-1 can specially appear in tumor tissue and can be detected abnormally expressing in the bronchogenic carcinoma even in earlier period. At present,domestic researches about the expression of Y box-binding protein-1 in lung cancer celles and the distinction of the expression in different lung cancer celles are few.Objective: To analyse the expression of Y box-binding protein-1 in lung cancer cell and the distinction of expression in different lung cancer celles,and to approach that Y box-binding protein-1 as an new oncogene and new tumor market can be used as molecular therapy target point in bronchogenic carcinoma.Methods: Total RNA was extracted from the lung cancer cells and lung tissue,and YB-1 cDNA was gotten by means of RT-PCR,then the results were analyzed by signature potency ratio of YB-1andβ-actin. Secondly, total proteinum was extracted from the lung cancer cells and lung tissue, and YB-1 protein was detected by means of western blotting, also the results were analyzed by signature potency ratio of YB-1 andβ-actin. Finally, the contents of DNA were detected by flow cytometer.Then we analyzed the reference among the contents of DNA and the differentiations and metachoresises of lung cancer.Results: The expressions of YB-1 mRNA and Y box-binding protein-1 in these lung cancer cells are much higher than in normal lung tissues.And there are distinctions in expression level of these lung cancer cells.Together with the detection results of DNA, there are associations among 95-D cell, NCI-H446 cell and SPC-A-1 cell.That means there are statistics distinctions in the expression level of YB-1 mRNA and Y box-binding protein-1 and DNA contents(95-D>NCI-H446,NCI-H446>SPC-A-1).Conclusion: Y box-binding protein-1 may be a new tumor marker of lung cancer. And there is a reference among Y box-binding protein-1 and the differentiations and metachoresises of lung cancer. It can be a new molecular therapy target of lung cancer.
