Study On The Anti-Tumor And Immune Effects Of SXKA Granules

Background: Malignant tumor is one of the most serious diseases in human body. How to prevent and cure tumor become the most important task, which draw general attention in the medical field. Up to the present day,the treatment of the malignant tumor usually adopt surgical removal, chemotherapy and radiation therapy.But those therapies can not cure the tumor,and the prices of medical treatment are too expensive for tumor sufferers.SXKA Granules is a chinese traditional medical prescription in clinic. SXKA Granules are used clinically to cure esophagus cancer, gastric cancer, bladder carcinoma,etc, with good results,here we test it in experimental tumors.Objective: To investigate the effects of SXKA Granules in experimental mice tumors and their action, on mice immune systems were observed .Methods:⑴Anticancer activities were tested in mice experimental tumor (H22,S180,EAC,Lewis and W256), the tumor bearing mice were randomly divided into control group, cyclophosphamide group 25 mg/kg and SXKA Granules 20,10,5 g/kg groups, and were treated for 10 days each,treated solid tumors were weighed respectively to observe its antitumor effects,and the inhibition rates on H22,S180,EAC,Lewis and W256 translanted tumors were calculated.⑵The survival times were tested in experimental mice tumor (P388), the tumor bearing mice were randomly divided into control group, PDD group 2 mg/kg and SXKA Granules 20,10,5 g/kg groups, and were treated for 10 days each, followed by observing and registering the survival time of mice.The rate of life prolongation of the P388-bearing mice were calculated.⑶To test the effect of SXKA Granules on the immune organ of S180-bearing mice . S180-bearing mice were sacrificed ,mice thymus and spleen were cut and weighed ,spleen index and thymus index were calculated.⑷The S180-bearing mice immunized with chicken red blood cell were randomly divided into tumor bearing group,cyclophosphamide group 25 mg/kg and SXKA Granules 20,10,5 g/kg groups, and another normal mice were used as control group.Adopt determinate method of hemolysin to test mouse serum half hemolysis value , to evaluate the actions of SXKA Granules on mouse humoral immunity system.⑸Adopt carbon clearance assay to test the phagocytosis of macrophage of the reticuloendothelial system(RES)on S180-bearing mice to evaluate the actions of SXKA Granules on mice cellular immunity system.Results:⑴SXKA Granules 20,10 g/kg dose groups had significant anti-tumor activity on H22,S180,EAC,Lewis carcinoma of mice and bearing W256 of rats.Compared with control,①the mean tumor weight of H22 of SXKA Granules three dose groups were decreased significantly(P<0.01,P<0.05),and the mean inhibition rates of SXKA Granules 20,10 g/kg dose groups were above 30%;②the mean tumor weight of S180 of SXKA Granules three dose groups were decreased significantly(P<0.01),and the mean inhibition rates of SXKA Granules 20,10 g/kg dose groups were above 32%;③the mean tumor weight of EAC of SXKA Granules three dose groups were decreased significantly ( P<0.01, P<0.05),and the mean inhibition rates of SXKA Granules 20 g/kg dose groups were above 38%;④the mean tumor weight of Lewis carcinoma of SXKA Granules three dose groups were decreased significantly(P<0.01, P<0.05),and the mean inhibition rates of SXKA Granules 20,10,5 g/kg dose groups were above 36%;⑤the mean tumor weight of W256 of SXKA Granules three dose groups were decreased significantly ( P<0.01, P<0.05),and the mean inhibition rates of SXKA Granules 20,10 g/kg dose groups were above 32%;⑵Compared with control,SXKA Granules 20,10 g/kg dose groups had extended the survial time of the P388-bearing mice respectively(P<0.01),and the mean prolong rates of SXKA Granules 20,10 g/kg dose groups were above 50%;⑶Compared with S180-bearing group, SXKA Granules 20,10 g/kg dose groups could increase the weight of thymus and spleen, Spleen index and thymus index were increased, SXKA Granules 5 g/kg dose group could increase thymus index(P<0.05);⑷As Compared with control group, SXKA Granules 20,10 g/kg dose groups could improve mouse serum half hemolysis value depressed by transplanted tumor dramatically(P<0.01), which revealed the SXKA granules could improve the mouse humoral immunity system;⑸SXKA Granules 20 g/kg dose group could increase of englobe indexαon S180-bearing mice remarkably(P<0.01), which indicated the SXKA Granules could improve their cellular immunity system.Conclusing: The results showed SXKA Granules that has obvious antitumor effects,and extend the survial time of the P388-bearing mice respectively,protect the immune organ,improve the mouse humoral immunity system and cellular immunity system remarkably.
