Traditional Chinese medicine compound prescription(TCMP)is the selection of appropriate Chinese medicine based on the principle of monarch,minister,assistant and envoy.The chemical compositions are complicated,and TCMP have multiple metabolic pathways and may act on multi-targets to play a role in clinical treatment.Effective quality control and comprehensive evaluation of its efficacy have become the focus of discussion in recent years.Research on the effective component combination and mechanism of compound Chinese medicine is a breakthrough point in the modernization of traditional Chinese medicine.Explaining the mechanism of multi-components and multi-target of TCMP from the perspective of modern science,we can better understand the basis of the efficacy of traditional Chinese medicine.Fupo Ganmao(FPGM)Granules are widely used in clinic,but there are few studies on the material basis and active mechanism.Based on the technology of liquid chromatography-mass spectrometry(LC-MS),the extraction solution of RAW264.7 mouse macrophages and FPGM Granules were used as the research objects,the potential active components were quickly identified by exploring the intracellular components.Furthermore,the toxicity,anti-inflammatory activity and anti-cancer activity indexes of the monomers and compositions of the potential active ingredients were discussed,and the compatibility rule of the active ingredients and the possible active mechanism were clarified.The main contents and results were described as follows:1.Screening and quantitative study of potential active components of FPGM Granules.By the use of LC-MS,this study used the extraction solution of RAW264.7 mouse macrophages and FPGM Granules as research targets,establishing methods to identify six potential components(hesperidin,arctiin,arctigenin,nobiletin,honokiol and magnolol).Meanwhile,the content of six different batches of preparations were detected individually.The results show that the accuracy,precision and stability of the established quantitative method are all in line with the requirements.The method is accurate,sensitive and reliable,which can provide reference for the multi index quality control of the preparation.2.The study of the anti-inflammatory activity of active constituents of FPGM Granules.Taking RAW264.7,a mouse macrophage induced by LPS,as an anti-inflammatory model in vitro,and six potential active monomers screened by high throughput as the main research objects,MTT method was used to determine the drug safety concentration.Griess method was used to detect the effect of monomers on NO secretion of RAW264.7 cells stimulated by LPS.In order to further study the effect of multiple compounds on NO secretion,explore the relationship between compounds,and to verify the results,other cytokines(TNF-? and IL-6)were detected by ELISA.3.The study of the anti-tumor activity of main active components of FPGM Granules.The A549 cell line was used as the tumor model to determine the effect of different drug concentrations on the survival rate of A549 cells by MTT.The results showed that the FPGM Granules,three monomer compounds and the combination group could inhibit the growth of lung cancer A549 cells in vitro.
