| ObjectiveTo investigate the protective effect of DAG oil in rats from non-alcoholic liver disease compared with TAG oil.Try to find out DAG oil's mechanism of action on the protection of NAFL.To expand DAG's function on medicine.Methods45 male SD rats (the average weight was 130±10 g )were randomly divided into 3 groups(10,17,18 separately) according to their weight. The three groups were normal- group(NG), diacylglycerol group (DAG)and triglyeride group(TAG). Animal feeds were prepared by ourselves according to AOAC prescription.The energy provided by fat in each group is 18% , 38.6% and 38.6% separately.We kept the quantity of food taken in each group equal.Except that,rats could eat and drink freely.Blood was taken from eye sockets every four weeks for certain index..At the end of the experiment, the fasting liver,fat and blood samples were collected,light microscopic observed the histology of liver with HE dying and analyzed including ALT, AST, TC, TG, TBA, ALP, GGT, HDL, LDL, SOD, MDA, CHE, TNF-α. All data were expressed as means±SD, and the analysis were performed with SPSS 11.0 for windows.Results1,The weight of rats in TAG group increased obviously compared to the normal group, the same as perirenal fat and testicle fat.2,The blood glucose in DAG group was lower than the glucose in TAG and normal group.3,Light microscopic observed the histology of liver with HE dying:the typical hepatic injury change of chronic alcoholism were observed in rats of TAG group.No obvious changes were found in DAG group.The rats of normal group showed the normal histology of liver.4,The ALT, AST, TC and TG level in serum of TAG group were significantly increased compared with the normal group. There was no significant difference of ALT, AST, TC and TG level in serum between the DAG group and the normal group.5,The serum SOD of the TAG group was lower and the serum MDA was higher compared with the normal group.The serum SOD of the DAG group was significantly higher compared with the TAG group.The serum MDA of the DAG group was lower than that of the TAG group.The serum CHE of the TAG group was higher compared with the normal group and the DAG group.There was no significantly difference in serum CHE level between the DAG group and the normal group.6,The serum TNF-αof the TAG group was significantly higher compared with the normal group. The serum TNF-αof the DAG group was decreased compared with the TAG group. There was no significantly difference between the DAG group and the normal group.Conclusions1,DAG might protect rats from non-alcoholic liver disease.2,DAG might increase the antioxidant ability in rats,such as SOD.3,DAG might restrain the secretion of TNF-α.4,DAG might reduce the leptin level,and it has no function on leptin resistence. |
