Objective To Study of diacylglycerol's effect on the blood fat and antioxidation of rats.Methods 60 Sprague-Dawley rats of 4 weeks old were randomly divided into five groups, 12 per group, half of male and female. Animal feeds were prepared by ourselves according to AIN-93G prescription.100% triglyceride group (100TG) was fed with high fat diet which contains 20% TG, while the 20% oil in feeds of 80% diacylglycerol group (80DG), 50% diacylglycerol group (50DG) and 30% diacylglycerol group (30DG) contain 80%DG, 50%DG and 30%DG respectively. The normal control group was fed with basal diets which contains 7% TG The energy provided by fat in triglyceride and diacylglycerol group is 40.4%, while normal control group is 15.6%. The rats were sacrificed under anesthesia after feeding of 8 weeks; compared the weight change between each group. The rats were sacrificed under anesthesia after feeding of 8 weeks, collecting adipose tissue for bady fat ratio and Lee's index; Collecting the liver for the study of histopathology; Detecting Alanine aminotransferase (ALT), aspartate aminotransferase (AST), TG and total cholesterol (T-CHO) of serum for liver function and lipid metabolism level evaluation; Detecting superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) content of serum for antioxidant system function evaluation.Results HE staining showed that the hepatic steatosis in 100TG group was higher than DG groups. Different degree of Inflammatory infiltrate was found, but no fibrosis lesions.The activity of AST in 80DG group was 78.20U/L, which was lower than TG Group (109.5U/L) (P<0.05). The Lee's index, fat percentage and blood fat in groups with additives of diacylglycerol were lower than TG group with statistics significance (P<0.05). The activity of superoxide dismutase in 80G group was 244.35U/ml. The activity of glutathione peroxidase in 80DG and 50DG groups were 1679.03 U/ml and 1479.55 U/ml respectively. The serum malonyldiadehyde in 80DG,50DG and 30DG groups were 3.66 nmol/ml,5.70 nmol/ml and 6.39nmol/ml respectively. The results of activity of superoxide dismutase, activity of glutathione peroxidase and serum malonyldiadehyde in TG group were 204.82U/ml,1106.93U/ml and 10.09nmol/ml respectively, these differences in the aspects had statistics significance (P<0.05).Conclusion Diacylglycerol could increase the anti-oxidative capacity, adjust the blood lipid metabolism, lower the adipose generation and ameliorate blood fat and oxidative stress in rats compared with triglyceride, but it needs more evidence to be confirmed. Appropriate dosage of DG of weight control should be further explored to provide more effective safe replacement to market.
