Objactive To construct tissue microarrays. To study the expression of APE/ref-1 in lung cancer, investigate the relationship between APE/ref-1and lung cancer. Methods Collect cases of paraffin-embedded specimen,and construct tissue microarrays.The immunohistochemical stain methods was used to detect the expression of APE/ref-1 protein in tissues. Results 1. Succeed in constructing 3 blocks of tissue microarray. They were answered for the request of research. 2. Protein expression level and the cellular location of APE/Ref-1 was different in lung cancer ,paratumour tissues and normal tissues. The positive rate of APE/Ref-1 was 68.02%(134/197) in lung caner tissues,27.08% (13/48) in the paratumour tissues and only 5%(1/20) in normal lung tissues.Their difference was significant(P<0.05). The shifts of APE/Ref-1 from nucleus to cytoplasm in lung cancer tissue was 8.63(17/197), the paratumour tissues was 0%,Their difference was significant(P<0.05)3. The expression level of APE/ Ref-1 was related to tumour stages,cell differentiation, lymphnode involvement and distant metastasis.4. 3-year survival rate of APE/ Ref-1 in positive group and negative group was 28.57% and 52.50%,the difference between the two groups statistically significant (P<0.05). Conclusions: 1. Tissue microarray can provide a large scale of immunohistochemistry detection of lung cancer in a short period with efficiency. 2. Over expression of APE/ Ref-1 protein relate closely with the carcinogenesis and progress of lung cancer. 3. Over expression of APE/Ref-1 or shift of APE/Ref-1 protein from nucleus to cytoplasm was related to the poor prognosis of patients with lung cancer.
