Study On The Changes Of Immunity Function And Spermatogenesis In The Model Mice With The Kidney-yang Deficiency

Objective To observe the change of the immunity function and spermatogenesis in the model mice with the kidney-yang deficiency and the effect of JinGuiShenQi Wan.Methods 60 healthy male mice of Kunming species were used, and randomly divided into 3 groups: contrast group, model group, treated group with JinGui ShenQi Wan. There were 20 mice in each group. Kidney-yang deficiency male mice model were induced by forcing mice to swim to make them over-fatigue and inducing male mice to sexual strain. It is by using immunohistochemistry method to measure the thymus index, spleen index and the changes of IL-2.The concentration of testosterone was examined by using radioimmunoassay method. Convoluted seminiferous tubules in testicle and the number of spermatogenesis cells were observed. To evaluate testis spermatogenesis function by using of Johnsen's quantitative measurement chart. The expression of Bcl-2 in testicle was examined with immunohistochemistry. Results The male mouse model is successfully established with excess sexual activity and over fatigue. The thymus index,spleen index and the expression of IL-2 in kidney-yang deficiency male mice decreased remarkablely compared with the control group (P<0.05) .The number of seminiferous tubule in testicle, number of cell of producing sperm, the concentration of testosterone and the expression of Bcl-2 in testicle in kidney-yang deficiency male mice decreased. .All those datum showed some improving tendency after treating with JinGui ShenQi Wan compared with the model group. Conclusions The male mouse model can be successfully established with excess sexual activity and over fatigue. The immunity function of kidney-yang deficiency male mice decreased evidently, and spermatogenesis was damaged. Formula tablet notifying the kidney can improve the immunity and procreation.
