Objective:Using intervention in vivo of rat to study the mechanisms of taurine(Tau)preventive neurotoxicity induced by manganese(Mn)in rat's prefrontal cortex and hippocampus.Methods:SD rats were divided into four groups after one week of observation:normal control,the group animals received daily of intraperitoneal(ip.)injections of sterile saline for 12 weeks; Mn treated group(Mn):rats were received ip.injection of MnC12.4H20 once a day for 12 weeks;Tau preventive group(Mn+Tau):The Mn level of this group are the same as Mn's,the Tau level is 200mg/Kg,three times per week,the assayes remained for 12 weeks;Tau treated group(Mn→Tau):After received the daily injection of Mn as Mn group for 12 weeks,the rats received Tau three times per week for 12 weeks.The dose Of Mn and Tau were the same as above, the experiment lasted 24 weeks.At the end of the experiment,all rats were decapitated,their striatum were removed and prefrontal cortex and hippocampus were abruptioed.Detecting indexes as flowing:the morphological changes of neurons,the apoptotic morphology and Apoptosis ratio in neurons,activity of SOD and productions of MDA,the protein expressions of caspase-3,bax and bcl-2 in brain.Results:①Mn induced apoptosis of neurons in rat's prefrontal cortex and hippocampus.②Mn inhibited activity of SOD and increased the production of MDA and Tau has the reverse effect.③Tau decreased level of apoptosis of neurons that caused by manganese.④Mn induced caspase-3 and Bax,inhibited Bcl-2 in neurons,besides,Tau induced Bcl-2 overexpress and raise the ratio of Bcl-2/Bax.conclusions:Mn induces Apoptosis in rat's prefrontal cortex and hippocampus neurons.The main mechanisms of Tau preventing cytoxicity against Mn were of reduce the oxidative stress and its effective control of the expressions of apoptotic factors in prefrontal cortex and hippocampus neurons.
