Objective To investigate the protection of trehalose on vascularized allogeneic bone after cryopresservation, and observe the patency of the pedicle and the osteosis of bone allografts, and find out the diference of protection between trehalose and other protective additive.Methods 36 New Zealand rabbits divided into 3 groups were evaluated. 12 rabbits were donors, 24 rabbits were hosts. Allotransplantation of cryopreserved radius diaphysis with the vascular pedicles which used trehalose as protective additive were implanted to 8 rabbits and immunosuppressive agent(CsA 10mg/kg/d) was administrated postoperatively for 4 weeks (group A ): Allotransplantation of cryopreserved radius diaphysis with the vascular pedicleswhich used dimethyl sulfoxide(DMSO) as protective additive were implanted to 8 rabbits and immunosuppressive agent(CsA 10mg/kg/d) was administrated postoperatively for 4 weeks (group B); Allotransplantation of cryopreserved radius diaphysis with the vascular pedicleswhich used trehalose and dimethyl sulfoxide(DMSO) as protective additive were implanted to 8 rabbits and immunosuppressive agent(CsA 10mg/kg/d) was administrated postoperatively for 4 weeks (group C). Radiograph was taken for each graft at 2, 4, 8, 12 weeks. 12 weeks after operation, the patency of the pedicle was observed through ink infusing and histological studies were done, and bone density was taken at 12 weeks. In addition, we take some bone from the three group to make histological observe and electron microscope observe l week after cryopreservation.Results To study the patency rate, bone density, the osteosis of bone allografts, the histological examination, the electron microscope examination, group A, C was superior to group B obviously, and group C was superior to group A. To study the bone density, group A was superior to group B obviously, group C was superior to group B obviously, group C was superior to group A obviously.Conclusions Using trehalose as protective additive of cryopreservation, assures the patency rate, speeds up the osteosis of bone allografts, and cytoactive of bone.
