Objective:This study aims to test Ezrin and AKT2 expressions in colorectal carcinoma, adjacent normal colorectal mucosa and combine with some clinical pathological factors to discuss the relationship between expressions of Ezrin and AKT2 and the progress, metastasis and prognosis of colorectal carcinoma.Methods:Immunohistological Elivision staining was used to detect the expressions of Ezrin and AKT2 in 95 cases of human colorectal carcinoma and 40 adjacent normal colorectal mucosa and the different expressions of Ezrin and AKT2 among lymph nodes metastatic and non-metastatic human colorectal carcinoma,including 54 metastatic cases and 41 non-metastatic cases were also under investigation.The relationship of Ezrin and AKT2 expressions with clinicopathological factors in colorectal carcinoma was also analyzed.Results:In adjacent normal colorectal mucosa,the expression of Ezrin protein was found in 4 cases among 40 cases(positive rate 10%).The expression of AKT2 was found in 7 cases among 40 cases(positive rate 17.5%).In colorectal carcinoma, the expression of Ezrin protein was found in 74 cases among 95 cases(positive rate 77.9%). the expression of AKT2 was found in 55 cases among 95 cases(positive rate 57.9%). the expressions of Ezrin and AKT2 have close relation to lymph node metastasis.The positive rates of Ezrin and AKT2 expressions in lymph node metastasis cases were obviously higher than those without lymph node metastasis. the expression of Ezrin protein in AKT2 positive cases was obviously higher than those of AKT2 negative cases.The lymph node metastatic rate was 61.2% in 49 patients with both positive expression of Ezrin and AKT2,while none of lymph node metastasis were found in the 16 cases with both negative.Conclusions:1.The increase in expression of Ezrin was closely correlated to metastasis in human colorectal carcinoma.It might be served as an important factor for determining tumor biological behavior and evaluating prognosis and metastasis.2.The high positive rate in colorectal cancer of AKT2 may be present in colorectal cancer and AKT2 may be the predominant isoform of AKT involved in carcinogenesis and development of colorectal cancer.The expression of AKT2 is closely related to lymph node metastatis,and it could be used to evaluate prognosis.3.Positive correlations were observed between the expression of Ezrin and AKT2 in human colorectal carcinoma. Combinational examination can evaluate prognosis and metastasis.
