AIM:The immunosuppressed mice were induced by cyclophosphamide and immunological liver injury mice was induced by Bacillus Calmette Guerin(BCG) plus lipopolysaccharide(LPS),which were used to investigate the immunomodulatory effects of qishile(QSL) and its mechanism.METHODS AND RESULTS:1.The effects of QSL on the immunosuppressed mice were induced by cyclophosphamideThe immunosuppressed mice were induced by cyclophosphamide(Cy,ip.). Immune organs were weighed and index of immune organs were calculated. Phagocytosis of mononuclear macrophage was determined by cleaning carbon particle method.Spectrophotography was used to estimate levels of serum hemolysis IgG and IgM.Plaque forming cell were determined with QHS.The mouse DTH model induced by DNFB was used to study the effects of hesperidin on the levels of DTH and subset of T lymphocyte.QSL(450,900,1800mg/kg,ig) could significantly increased K,a number,the hemolysin level in splenocyte and IgM,IgG in serum.Index of spleen and thymus,were increased at the dose of 900 and 1800mg/kg.In addition,QSL(450,900,1800mg/kg, ig) could regulate DTH response to normal level.Meanwhile,QSL significantly increased both CD4~+ and CD8~+ cell counts,especially influencing the number of CD4~+ cell at the dose of 450,900,1800mg/kg.The results that showed QSL have an enhanced effect on immunosuppressed mice mainly by improving nonspecific immunity,humoral immunity and cell immunity. 2.The protective effects and its mechanism of QSL in immunological liver injury miceBacillus Calmette Guerin(BCG) and lipopolysaccharide(LPS) from tail vein were used to induce immunological liver injury in mice.Index of liver and spleen were tested.The histopathologic changes in the liver of mice were observed. Meanwhile the alanine aminotransferase(ALT),aspartate aminotransferase(AST) and nitric oxide(NO) levels in serum and alondiadehyde(MDA) content,superoxide dismutase(SOD) activities in liver homogenate were assayed by spectrophotometry. TNF-a and NF-κBp65 in hepatic tissues were determined by the method of immunohistochemistry,the levels of iNOS mRNA expression in liver were tested by reverse transcription polymerase chainreaztion(RT-PCR).QSL(900,1800mg/kg,ig ) could decrease the enlarged liver,spleen index in mice.QSL(900,1800mg/kg,ig ) were found to significantly decrease the serum transaminase activities.Meanwhile,QSL also decreased MDA,iNOS,NO contents and prevented the reduction of SOD activity in liver homogenate.QSL(450,900, 1800mg/kg,ig) could inhibit the expression of TNF-a,NF-κB p65 and iNOS mRNA in liver tissue.The results showed that QSL have significant protective action on mice immunological liver injury.Its mechanism might be related to depleting ORF, suppressing cell lipid peroxidation and accommodating the path of NF-κB,diminishing mediators of inflammation.
