Objective: To explore essence of hyperthyroidism with hyperactivity of liver-yang by proteomics method to establish two-dimensional electrophoresis profiles from hypothalamuses and adrenal glands of hyperthyroidism rats with hyperactivity of liver-yang and normal rats, to analysis the different expression proteins of the two kind rats, and to research function and effect of those proteins.Methods: (1)20 Spraguedawlay rats were randomly divided into normal control group (normal group, n=10) and model control group (model group, n=10). Normal group rats weren't treated, and had normal food and drink. Model group rats were administered Guizi Decoction and L-T4. So an animal model of hyperthyroidism with hyperactivity of liver-yang was established. All rats were killed on 22th day. General information and T3,T4 were indexes, these indexes decided whether succeeded to copy an animal model.(2) Hypothalamuses and adrenal glands of the two groups were directly sampled for proteomic analysis.Results: For hypothalamuses tissues in normal rats, a total of 764±30 spots were detected. For hypothalamuses tissues in model group, a total of 755±28 spots were detected. For adrenal glands tissues in normal rats, a total of 690±33 spots were detected. For adrenal glands tissues in model group, a total of 715±25 spots were detected. In hypothalamuses tissues 18 proteins increased and 24 proteins decreased in model group, compared with normal group, including Thioredoxin, Ubiquitin carboxyl-terminal hydrolase isozyme L1, Platelet-activating factor acetylhydrolase IB gamma subunit, glutamate dehydrogenase, dihydropyrimidinase-related protein 2, NSFL 1 cofactor p47, Tubulin beta 5. In glands tissues 33 proteins increased and 6 proteins decreased in model group, compared with normal group, including GrpE protein homolog 1, 60kDa heat shock protein, calreticulin precursor, Acyl-Coenzyme A dehydrogenase, Isovaleryl-CoA dehydrogenase and Aldehyde dehydrogenase.Conclusion: (1) We established the expression maps of hypothalamuses and adrenal glands tissues in hyperthyroidism with model and normal rats.(2) It is different for the expression proteins of 2-DE profiles of hypothalamuses and adrenal glands tissue in normal group and model group.(3)Using mass spectra technology, 13 differential proteins were identified. These differential proteins might take part in formation of hyperthyroidism with hyperactivity of liver-yang by oxidation-reduction reaction, path of proteinaceous degradation, cell division and signal transmit, but specific mechanism required further study. It provides new thinking for hyperthyroidism therapy and diagnoses.
