ObjectiveTo investigate the expression of membrane-type 1 matrix metalloproteinase protein (MT1-MMP) and micro vessel density (MVD) in human endometrial carcinoma tissues and their relations with the invasion and metastasis of endometrial carcinoma..MethodsImmunohistochemical staining (SP method) was used to detect the expression of MT1-MMP protein and MVD in endometrial carcinoma tissue of 48 patients,the non-typical proliferation in endometrium of 10 samples and the normal endometrium during the period of proliferation of 30 samples.Results1.The positive expression proportions of MT1-MMP in endometrial carcinoma tissues were higher than those in the non-typical proliferation (P<0.05) and in normal endometrial samples (P<0.01). But it did not show different expression with the presence of surgical pathologic stagingⅠ-ⅡandⅢ-Ⅳ(P>0.05).There was no statistical significance with the histologic grade(G1,G2 and G3) (P>0.05).In different depth of myometrium invasion, there was obvious difference in expression of MT1-MMP (P<0.01). There was statistical significance between endometrial carcinoma with lymphatic metastasis and without lymphatic metastasis in expression of MT1-MMP (P<0.05).2.MVD in endometrial carcinoma tissues was higher than those in normal endometrial samples. There was obvious significant (P<0.05). MVD in surgical pathologic stagingⅠ-ⅡandⅢ-Ⅳwas no obvious difference (P>0.05), MVD in G1,G2 and G3 was no obvious significant (P>0.05). MVD staining scores in tumor cells was also significantly associated with the depth of myometrial invasion (P<0.05). MVD staining scores in tumor cells was statistical significance between endometrial carcinoma with lymphatic metastasis and without lymphatic metastasis (P<0.05).3.MVD in tumor cells was obvious statistical significance (P<0.05) between MT1-MMP (+) and MT1-MMP (-).Conclusions1. The MT1-MMP expressed in normal endometrial samples,non-typical proliferation and endometrial carcinoma. The positive expression proportions of MT1-MMP in endometrial carcinoma tissues were higher than those in the non-typical proliferation (P<0.05) and in normal endometrial samples (P<0.01), which suggests that the expression of MT1-MMP was connected with endometrial carcinoma.2. The expression of MT1-MMP in endometrial carcinoma was obvious increased with the increasing invasion depths (P<0.01), the positive express proportions of MT1-MMP in endometrial carcinoma tissues was higher in lymphatic metastasis than those in without lymphatic metastasis(P<0.05),which suggests that the positive express proportions of MT1-MMP was related to the tumor invasion and metastasis of endometrial carcinoma.3. MVD in tumor cells was higher than those in normal endometrial samples (P<0.05) and the non-typical proliferation (P>0.05). MVD in tumor cells was also significantly associated with the depth of myometrial invasion (P<0.05). MVD in tumor cells was statistical significance between endometrial carcinoma with lymphatic metastasis and without lymphatic metastasis (P<0.05).It showed that The increasing expression of MVD in tumor cells may be associated with the depth of myometrical invasion, which suggests that angiogenesis plays an important role in invasion and metastasis of endometrial carcinoma.4. MVD in tumor cells was obvious statistical significance between MT1-MMP (+) and MT1-MMP (-). MT1-MMP may accelerate the tumor invasion and metastasis in endometrial carcinoma.
