Study On The Correlation Between Hypoalbuminemia And TNF-alpha, IL-6 In Advanced Colorectal Cancer Patients | | Posted on:2009-06-03 | Degree:Master | Type:Thesis | | Country:China | Candidate:H J Song | Full Text:PDF | | GTID:2144360245953397 | Subject:Oncology | | Abstract/Summary: | PDF Full Text Request | | Objective To explore the relationship between hypoalbuminemia and the level of TNF-αand IL-6 in advanced colorectal cancer patients,through testing the levels of TNF-αand IL-6 in serum,and the expression in the cancer tissue.Methods According to the circulating levels of albumin,59 patients with advanced colorectal cancer were divided into 2 groups:35 patients with hypoalbuminemia(albumin≤35g/L)classified as group A,and other 24 patients (albumin≤35g/L)as group B.1.The levels of TNF-αand IL-6 in patients' sera were analyzed by enzyme-linked immunosorbent assay(Elisa testing),and 15 normal examinees were selected as controls.Experiments were operated accordancing with the specification strictly,then the OD450nm of standard and each sample was measured,and density was calculated by the standard curves.2.Immunohistochemistry testing(SupervisionTM)was used to detect the expression of TNF-αand IL-6 in cancer tissues in each groups above -mentioned.Adjacent normal tissues(pathological examination:noncancerous tissue)of each case were selected as controls. Results Part 1:Compared with the normal controls,the levels of TNF-αand IL-6 in advanced colorectal cancer patients was increased significantly (P=0.001,P=0.034).The level of TNF-αand IL-6 in group Awas significantly higher than that in group B(P=0.000,P=0.002).Significantly,the circulating level of albumin was negatively correlated with TNF-αand IL-6 in serum (Alb vs.TNF-α:r=-0.606,P=0.000;Alb vs.IL-6:r=-0.439,P=0.001).Part 2:In cancer tissues of all 59 cases,both the expression of TNF-a and IL-6 were uncorrelated with the Dukes stage(P=0.386,P=0.594),there was no significant difference between group A and group B(P=0.275)of TNF-a immunohistochemistry,while IL-6 immunohistochemistry were significantly different(P=0.006).In the adjacent normal tissues,two cases were only seen exhibiting faint(+)IL-6 immunohistochemistry,and one case was seen expressing(+)of TNF-a.The degree of lymphocytes and macrophages infiltrating in the interstitial tissue in group A were stronger than these in group B(P=0.030).Conclusion①Hypoalbuminemia is associated with the increasing of TNF-αand IL-6 in advanced colorectal cancer patients.The interaction of cancer tissues and host is possibly the important reason of the occurrence of hypoalbuminemia.②Consequently,TNF-αand IL-6 levels can be used as new basilic targets to treat hypoalbuminemia in advanced colorectal cancer patients. | | Keywords/Search Tags: | Colorectal cancer, Hypoalbuminemia, Albumin, Tumor necrosis factor-α, Interleukin-6 | PDF Full Text Request | Related items |
