Objective: To culture rat ovarian granular cells and osteoblasts in vitro, this exprement is observed the effects of abstractive parts of Duzhong leaves and its components on the estrogen production in cultured rat ovarian granular cells, which helps to probe into the mechanism that abstractive parts of Duzhong leaves prevents postmenopausal osteoporosis.Method: Rat ovarian granular cells were cultured in DMEM medium. After 24 hours, different concentrations of abstractive parts of Duzhong leaves were added to the medium and ovarian granular cells were cultured for 48 hours. Cells multiplication was measured by MTT. Estradiol in different medium were measured by Enzyme immunoassay. Primary osteoblastic cells were obstained from the calvaria of newborn SD rats by digestion with 0.1% collagenase and cultured in DMEM medium. Ovarian granular cells and osteoblasts with 2:1 and 1:2 were directly co-cultured for 72 hours. ALP in culture medium by osteoblast was tested.Results: After ovarian granular cells were cultured for 48 hours, abstractive parts of Duzhong leaves increased estradiol production. Compared with the blank group, abstractive parts of Duzhong leaves could promote the quantity of estradiol at a concentration 10-5g/ml (p<0.05). Compared with the osteoblast group, ALP state was increased in granular cells and osteoblast with 1:2 after 72 hours(p<0.05).Conclusion: The abstractive parts of Duzhong leaves stimulated production of rat ovarian granular cells in a dose-dependent fashion. To promote estradiol production is one of mechanisms of abstractive parts of Duzhong leaves in preventing postmenopausal osteoporosis.
