Experimental Study On The Ovarian Granular Cell Apoptosis And Inhibin α-subunit Expression In Female Rats At Menopausal Transition Period

Objective:To observe the female rats' ovarian follicular development,folliclular atresia and granular cell apoptosis in different stages of menopausal transition period ,then study the tendency of apoptosis-related gene bax ,bcl-2 expression and Inhibina-subunit in granular cell.Methods :10 months old,12 months old ,14 months old ,16 months old, 18 months old female rats without oestrum were chosen , every group had 8 rats. 2.Ovarian folliclular development ,folliclular atresia of different group were observed by light microscop and granular cell apoptosis were observed ultrastructure by transmission electron microscope .3.Observing expression tendency of Bcl-2 protein, Bax protein and Inhibin α-subunit in granular cells at menopausal transition period used immunohistochemistry technology . Results:1.There were many primary follicles ,secondary follicles , atresic follicles and luteums at 10 months old rats' ovarian cortex. With the rats grown, the growing follicle and luteum decrease ,while the atresic follicles increase. And the ovarian cortex atrophy, almost no follicles developed in the last. 2.Through the transmission electron microscope it can be seen : In normal granular cell the nucleolus were clear-cut , euchromatin and heterochromatin distributed equal ,euchromatin content plenty, nuclear membrance integrity, smooth and glossy. And cytoplasm contained many mitochondria, rough endoplasmic reticulum, Golgi boby and other orhanells.In apoptosis earlier period ,there were edema and vacuolation in orhanells and mitochondrion swelling ,mitochondrial cristae break ever extinction in granular cell, while chromatin achieve nuclear membrance, nuclear membrance shrunken. Then the chromatin continue coahulation, shift to border. In apoptosis anaphase .granular cell nuclei stained deeper , cell conjugation scatter, and cell fragmenta ,apoptotic body can be seen.3.Immunohistochemistry: There were Bax protein ,Bcl-2 protein and Inhibina-subunit that expressed in ovarian granular...
