| Objiect:Since the relationship between the microsatellite instability(MSI) and hereditary nonpolyposis colorectal cancer(HNPCC) were found, there is still much debate on the distinct classes of genomic instability and their specific roles in the initiation of tumor formation. Microsatellite instability (MSI) and loss of heterozygosity (LOH), the alteration in length and strength of short tandem repeat sequences are an important molecular characteristic of many human tumors.MSI and LOH analysis has become an attractive method for diagnostic and tumor research purposes. In the reaserch,we investigate the relationship between the microsatellite instability(MSI),loss of heterozygosity (LOH) and breast cancer,bone tumor,to discover whether the microsatellite instability(MSI) and LOH play a important role in the development of this two tumors.We may use two metheods to discuss the result, and provide scientific theory for forensic identification.Methods : Twenty concer tissues taken form 20 different unrelated individuals and there bloodspecimens were examined with Chelex -100 and chloroform phenol extraction of DNA,then profiler plus PCR,we used polyacrylamide gel electrophoresis technique and 3100 Genetic Analyzer for analysis of amplification products.Results: Through the polyacrylamide gel electrophoresis technique anaIysis:MSI was the most frequently observed at D5S818 with an incidence of 25%.LOH at TH01 and D3S1358 each was detected in 5%(1/20) of the tumor;Based on the 3100 Genetic Analyzer method:Except two blood specimens haven't any amplification products,others had the amplification products.The genetic alterations occurred in 2 out of 20 cases. LOH of the bone tumor were both detected at D3S1358 and D13S317 in the number 6 tumor,the numbet 8 tumor at D8S1179 was generate LOH,moreover,at the Amel locus,we detected LOH.Conclusion : This study seems to show the evidence of TSG on D3S1358, D13S317andD8S1179 related to the bone tumor.The gene changes at D5S818 play a notabel role in the occurrence of the bone tumor.The function of the 3100 Genetic Analyzer method more stabilization, sensitiveness rather than polyacrylamide gel electrophoresis technique.The forensic community has to take be cautious not to use the tumor tissue for personnel identification. |
