| Objectives: To compare the results of proteomics between the donors and the hyperplastic thymus tissue from patients with myasthenia gravis .Methods: The proteins isolated from the hyperplastic thymus tissue from21 patients with myasthenia gravis (MG) and 11 donors were separated by techniques of 2-DE(Two-dimensional gel electrophoresis). The proteins were then stained with colloidal coomassie blue to produce a high-resolution map of the proteome. Selected proteins from this map were in-gel digested with trypsin and the resulting tryptic peptides were analyzed by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI -TOF-MS). Protein identification was finished by searching the PMF data in NCBInr protein data bank using Mascot program at http://www. matrixscience. com.Results : 10 abnormal protein spots were obtained. 8 of witch wereidentificated, 2 were unidentified. .There were 4 spots significantly increased, which are tubulin, beta Chain A, Thioredoxin Peroxidase B ER-60 protease Chain A, Crystal Structure Of The Ga Module Coraplexed With Human Serum Albumin: 4 spots significantly decreased, which are biliverdin reductase B Keratin, type I cytoskeletal 19 Chain B, T-To-T(High) Quaternary Transitions In Human Hemoglobin: Deshisl46beta Deoxy Low-Salt alloalbumin Venezia .Conclusions : These proteins from hyperplastic thymus tissue frompatients with myasthenia gravis (MG) were different of the donors.. It will be helpful to further research of MG and find new molecular markers for early diagnosis and treatment of MG. |
