Objective:In clinical for patients with primary hypertension were treated three skin buck soup, with a good antihypertensive effect and improve the r ole of clinical symptoms. We examined the effects of hypertension targeted herbs, of essential hypertension patients taking three skin treatment after the step-down soup, and out-patients were randomly selected using step-dow n proteomics approach for the three skin soup the basis for the treatment o f primary hypertension provide the scientific basis of the target protein m olecules antihypertensive medicine for treating hypertension in three batch es of blood pressure to provide a scientific basis.METHODS:A self-controlled, open study group.34 patients were enrolled in patients with essential hypertension, a high blood pressure in 8 cases,19 cases of grade 2 hypertension,7 cases of grade 3 hypertension. Significant differentially exp ressed proteins selected points, using matrix-assisted laser desorption ion ization time of flight mass spectrometry (MALDI-TOF-MS) and matrix-assisted laser desorption tandem time of flight mass spectrometry (MALDI-TOF/TOF-MS) Determination of peptide mass fingerprinting (PMF), Identification of pro tein database search peptide corresponding to the protein, to obtain inform ation on differentially expressed proteins.Results:1. Three skin buck Tang on blood pressure:three leather buck soup from the beginning the first week of taking blood pressure began to decrea se, blood pressure decreased significantly within 4 weeks,4 weeks stabiliz ed 4 weeks after the drug accumulated markedly, the effective total of 31 c ases, accounting for 91.18%. Accumulated markedly after 6 months, the effec tive total of 27 cases, accounting for 79.41%. Three step-down soup that sk in has a more long-term blood pressure, a stabilizing effect, blood pressur e drugs can be stable in the normal range.2. Three Buck Tang on skin symptoms, the improvement syndrome:cli nical essential hypertension in patients with vertigo, the first such bound, shortness of breath, fatigue, fatigue, greasy yellow tongue coating, slipp ery pulse string for the main symptoms.4 weeks after taking him for the me dical clinical symptoms had improved (P<0.05), total effective rate was 97. 06%, followed up for 6 months after the effective rate was 85.29%, showing the three Paper buck soup on a more significant improvement in symptoms, an d the efficacy and stability.3. The proteins identified are:haptoglobin-related protein precursor,α2 haptoglobin, human serum transferrin, apolipoprote in A1, turn thyroxine protein Thr119met, anti-inhibitor of apoptosis gene B cl-X1 And Bcl-2, keratin 10, heme binding protein, PR02044, hemoglobin, al bumin isoenzymes. These proteins are involved in cytoskeletal function, sig nal transduction, cell metabolism, electron transport and so on.Conclusion:1. Three skin soup can reduce blood pressure in patients with e ssential hypertension, blood pressure, and antihypertensive effect of a mor e lasting stability. The clinical symptoms were more specific effect.2. Application of two-dimensional gel electrophoresis and analy sis software for comparison of serum protein before and after treatment and found differences in 13 proteins points to prove three essential hypertens ion patients taking antihypertensive soup skin serum protein expression cha nged significantly. Differences in protein spots of 13, has by matrix assis ted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) and matrix-assisted laser desorption tandem time of flight mass spectr ometry (MALDI-TOF/TOF-MS) Determination of peptide mass fingerprinting (PM F),Identification of the peptide corresponding to the protein, access to th e 11 proteins differentially expressed points of information, from the prot ein level to explore the mechanisms of blood pressure medicine to a variety of protein factors may be involved.
