background: We evaluated biomicroscopic and histological effects on the anterior segment in the rabbit eye after temporary aqueous substitution with various amounts (0.15 ml and 0.02 ml) of perfluorodecaline(PFDL).To observe the effects of various amounts of perfluorodecalin on the anterior chamber angle and intraocular pressure in rabbits.Methods:0.15ml perfluorodecalin was injected into right anterior chamber of 16 white rabbits and the same does of balanced salt solusion was given to the controll group (4 rabbits),0.02ml perfluorodecalin was injected into right anterior chamber in an additional 4 white rabbits.The intraocular pressure was measured before and 1d,3d,1,2,3,4,5,6,7,8 weeks after the injection,The treatment rabbits were sacrificed .The eyes were enucleated and the histological examination was performed on the week of 3d,1,2,4,6and 8 postoperatively.Results:1.Perfluorodecalin, having a'fish eggs'or bubbles appearance,was noted in the inferior anterior chamber. There is associated intraocular inflammation in the anterior chamber.2.In three group ,the intraocular pressure(A teatment group:16.17±6.81;B teatment group:13.16±3.71;he controll group:12.12±4.22) is lowest on the first postoperative day,there's no statistic difference in three group(P<0.05).In A teatment group,A increment of intraocular pressure was measured at the week of 1 to 8 postoperatively(P<0.05), In B teatment group and the controll group, the intraocular pressure is similar to the peroperation level on the 3rd postoperative day.3.Histopathologically, A teatment group:Within the first 1 weeks,postoperatively corneal endothelial edema and degeneration ,time passing,, corneal endothelial cell loss was observed ,and new vessels emerge in corneal matrix.transparent single or fused vesicles and macrophages were accumulated at the anterior chamber angle in A treatment rabbits.some lymphocytes and neutrophils were aslo noted here.Trabeculae degenerated and inertrabecular space increased. conclusions:1. the anterior chamber injection of 0.15ml perfluorodecalin could lead to a longstanding increase of the intraocular pressure.2. 0.15ml perfluorodecalin is filled in the anterior chamber for three days, it could lead to reversible inflammatory reaction ;over this time, corneal and trabeculae degeneration were seen in the cornea the anterior chamber angle.3. 0.02ml perfluorodecalin is filled in the anterior chamber for 8 weeks, tissue damage is reversible , intraocular tolerance might be very close to 0.02ml.4. Mechanism of tissue damage on PFDL: (1) perfluorodecalin's specific gravity and mechanical barrier function; (2) perfluorodecalin's direct toxicity; (3) oxygen toxicity; (4) intraocular hypertension aggravates the damage of trabecular reticulum's cells.
