ABSTRACT:Heat shock proteins (HSP),also called stress protein(SP),are a sort of protective proteins, which are caused to produce in the cell or organs of creatures exposed to harmful conditions, such as being lack of blood or oxygen, heavy metal, gene damage, organism damage, bacterial infection and so on.In recent years, it causes widely interesting in field of sports biology. The studies what have done focuses on heart,skeletal muscle widely. Recently some scholars attempt to solve some problems concerning fatigue in training using the characteristics of heat shock proteins, and some successes achieved. Kidney is the organ of bodies whose rate of metabolism is the largest in bodies, which can show the conditions in bodies obviously. Now today it has been studied widely in field of physic. But most of these studies were concentrated on the ischemic reperfusion and damage caused by ischemic reperfusion,drug,pathology and so on of kidney, and caused heat response in podocyte of renal glomerulus,epithelium of renal capsule,epithelium of renal tubule et al. Furthermore, heat shock proteins induced by events above and protect against damage. The study of heat shock proteins in kidney in sports biology is scarce. Only a few articles concerning heat shock proteins expression of kidney.The objective of this study is to determine influences of different intensities on the expression of heat shock protein 70 in kidney in normal temperature after high temperature treatment. In the two different conditions, to observe the changes of antioxidant system in kidney at the same time, such as SOD,GSH-Px,MDA, and of CAT in serum. Furthermore, to detect the protection of heat shock protein 70 against response to running.Methode:3 months old male Sprague-Dawley rats, 48, were assigned 6 groups: sedentary control,15m/min group,27m/min group these are under normal condition and high temperature treatment groups such as sedentary group,15m/min,27m/mingroups these experienced high temperature treatment(42℃15min). The sedentary groups were killed in quiet, 15m/min and 27m/min groups all were killed immediately after running.Results:(1)The level of heat shock protein 70 in kidney was increased with the increase of intensities in both groups of normal temperature and high temperature treatment. The level of heat shock protein 70 in groups which were treated in high temperature was significantly higher than groups in normal temperature(P<0.05).(2)After the treatment of high temperature, the level of heat shock protein 70 in kidney increased rapider than in groups in normal temperature, the expression of heat shock protein70 mRNA in groups high treatment were rapider than in normal temperature groups.(3)After the treatment of high temperature, the level of malondialdehyde(MDA) in renal mitochondrion was decreased in groups of treatment in high temperature more than groups in normal conditions; Activity in group of 27m/min treated with high temperature was higher than in group of normal temperature(P<0.05).(4)After the treatment of high temperature, the activities of superoxide dismutase (SOD) in renal mitochondrion were higher in groups with treatment of high temperature than in groups in normal temperature.(5)With the intensities and load of running increasing, the activities of glutathione peroxidase (GSH-Px) were dropped in renal mitochondrion of rats in both groups of high and normal temperature. But the status of GSH-Px was higher in groups of 15m/min and 27m/min with high temperature treatment than in normal temperature groups, relevantly.(6)With the intensities and load of running increasing, the activities of catalase (CAT) increasing in blood serum, but in groups after high temperature treatment higher than in groups in normal temperature, relevantly.Conclusions:(1)The expression of heat shock protein 70 in kidney in group of 15m/min running over 60min in normal temperature immediately, which is to say that the intensity and load could elicit the expression of heat shock protein70 (HSP70) and elevate level of heat shock protein70.(2)The expression and level of heat shock protein70 in kidney in group of 27m/min running over 60min in normal temperature immediately higher significantly than sedentary control and 15m/min, which is to say that the expression and level of heat shock protein70 in kidney were increasing rapidly with the advance of intensity and load, furthermore, impact on kidney is very hard with intensity increasing.(3)After 24h high temperature(42℃15min)treatment, in 15m/min and 27m/min groups higher than sedentary control group which also experienced high temperature treatment in expression of heat shock protein70, and also higher than normal temperature groups relevantly, which determined that high temperature treatment could enhance expression and level of heat shock protein70 in kidney.(4)After high temperature treatment, the status of MDA in renal mitochondrion is lower than in normal temperature groups, however the activities of SOD and GSH-Px were higher than in groups in normal temperature, which determined the protective function of heat shock protein70 in kidney.(5)After high temperature treatment, the activities of CAT were higher in blood serum than in groups in normal temperature, which showed concentration of free radicals in blood serum, determined the protective function of heat shock protein70 in homeostasis in body.
