Purpose: To investigate the expression of TGFβ1 and NFκB in gastric carcinoma and precursor lesions, to study their relationship in growth progression of gastric carcinoma and clinical pathologic significance.Methods: For preparation of 109 of gastric adenocarcinomas, 28 of high grade dysplasia, 20 of low grade dysplasia, 30 of chronic atrophic gastritis, 29 of intestinal metaplasia and 21 of normal gastric mucosa tissue blocks, to made tissue micro-array. Immunohistochemistry was used to determine the distribution and intensity of TGFβ1 and NFκB staining in 109 of gastric adenocarcinomas, 28 of high grade dysplasia, 20 of low grade dysplasia, 30 of chronic atrophic gastritis, 29 of intestinal metaplasia and 21 of normal gastric mucosa, and to analysed the relationship of protein expression and clinical significance in gastric cancer. The results of immunohistochemical staining was to evaluate both the intensity of staining and the proportion of the stined epithelial cells. The percentage of positivity of the tumor cells and the staining intensity were then multiplied in order to generate the immunoreactivity score (IS) for each of the specimens.Results: The expression IS of TGFβ1 and NFκB in gastric adenocarcinomas and dysplasia (high and low grade) were significantly higher than chronic atrophic gastritis, intestinal metaplasia and normal gastric mucosa(p<0.001, p<0.001, p<0.001). Meanwhile, the expression of TGFβ1 was increased in advanced gastric cancer and clinical stageⅢ(p<0.01, p<0.05), and the expression of NFκB was increased in larger tumor and clinical stageⅣ(p<0.01, p<0.05). There was no significant correlation between the both expression of TGFβ1 and NFκB and patient 5 year survival. Positive relationship was shown between the expression of TGFβ1 and NFκB in gastric cancer (p<0.05).Conclusion: TGFβ1 and NFκB together take part in the form gastric tumor, and may have some relationship with gastric cancer invasion.
