Objective To investigate the clinical significance of p15 and transforming growth factor-β1(TGF-β1)expression in breast carcinoma as well as the relationship to each other between them. Methods the expressions of p15 and TGFβ1 in specimens of 62 cases of breast carcinomas, 15 cases of breast tissue surrounding carcinoma and 12 cases of normal breast tissue were studied by using SABC immunohistochemical staining. Results (1)The total positive rate of p15 protein expression in breast carcinomas was significantly lower (58.1%) than that in tissue surrounding carcinoma (93.3% P<0.05) and in normal breast tissue(100% P<0.05). The expression of p15 protein in breast carcinomas was not markedly related to patient's age, menopausal situation, tumor size, lymph node metastasis, histological grad, histological type, TNM stage, local recurrence, distant metastasis,ER, PR and prognosis (P>0.05). (2) The total positive rate of TGFβ1 expression in breast carcinomas was significantly lower (59.7%) than that in tissue surrounding carcinoma (86.7% P<0.05) and in normal breast tissue (91.7% P<0.05). The expression of TGFβ1 in breast carcinomas was showed to be highly related with lymph node metastasis, histological type, distant metastasis and ER, PR (P<0.01 P<0.05 P<0.001), but be not markedly associated with patient's age, menopausal situation, tumor size, histological grade,TNM stage and local recurrence (P>0.05). There was a inverse correlation between the extent of TGFβ1 positive expression and both lymph node metastasis and infiltration of breast carcinoma(P<0.01 P<0.05). The expression of TGFβ1 was significantly related with prognosis of the patients (P<0.01), and the prognosis of the patients with TGFβ1 positive tumors was markedly better than that of those with TGFβ1 negative tumors. (3) The positive rate of TGFβ1 expression was significantly higher in p15 positive breast carcinomas (80.1%) than in p15 negative those (30.8%)(P<0.05). Conclusions (1) Expression of p15 protein may be associated with breast carcinogenesis, but have no relationship with clinicopathological factors and prognosis of breast carcinoma. (2) TGFβ1 expression may be related to the growth, infiltration and metastasis of human primary breast carcinoma and serve as a marker to determining tumor infiltrative, metastatic potential and prognosis. (3) There is a significantly positive relationship between the expression of p15 and TGFβ1 in breast carcinoma.
