OBJECTIVE: To observe the effects of extracts from Inonotus obliquus onstreptozocin(STZ) induced diabetic rats and probe its mechanism and function onanti-diabetes. In the meantime, to provide the experimental evidence for Inonotusobliquus as a new kind of fungus drug which could prevent and cure diabetes.METHODS: A total of 8 healthy male Wistar rats were randomly chosen as controlgroup and the other rats were injected STZ (70mg/kg) into abdominal cavity to inducediabetic model. We would measure the levels of fasting blood glucose(FBG) after 6days.FBG≥16.7mmol/l suggested diabetic models were succesful. The diabetic ratswere randomly divided into 5 groups: diabetic model group,drug control group, lowdensity extracts from Inonotus obliquus group,middle density extracts from Inonotusobliquus group and high density extracts from Inonotus obliquus group. The druggroups rats were orally administered glucophage by 200mg/kg.Inonotus obliquusgroups were orally administered individually extracts from Inonotus obliquus by300mg/kg,600mg/kg and 900mg/kg.The normal control group and diabetic modlegroup were orally administered purified water by the same volume once a day for 40days. We should measure fasting blood glucose(FBG) of every groups before thetreatment and after. We measured the levels of glycosylated hemoglobin(GHb),serum total cholesterol(CHO),triglyceride(TG),low density lipoprotein,superoxidedismutase(SOD),malonal dehyde(MDA),alanine transaminase(ALT) andaspartate transaminase(AST) after experiment. In the meanwhile,we made use of HEstaining method, PAS staning method and immunohistochemistry staining method toobserve pathomorphology changes of pancreas,liver and nephros.RESULT: Extracts from Inonotus obliquus were able to degrade levels of bloodGHb and FBG and contents of serum CHO,TG,LDL-C and MDA significantly; They could advance SOD activity and degrade ALT and AST activity, and allexhibited concentration dependent relationship. Extracts from Inonotus obliquus wereable to make pancreatic island bigger and increase the number of B cell in pancreaticisland.They could increase hepatic glycogen and make glomcrulus smaller, what'smore,they could decrease the positive things of mesangial region.CONCLUSION: Extracts from Inonotus obliquus had effect of decreasing bloodglucose and regulating blood fat. They were able to resist oxidative stress and clear freeradicle. They could improve the utilization of glucose in liver and the function ofliver.They could repair the damage of pancreatic island and improve renal damage indiabetes. Effects of extracts from Inonotus obliquus in the treatment of STZ-induceddiabetic rats might be related with their polycose components decreasing blood glucoseand tannin resisting oxidative stress or clear free radicle.
