Objective To analyze protein expressions of MDM2 and PTEN in Hepatocellular Carcinoma(HCC), and to investigate relationships among MDM2 and PTEN and clinicopathologic factors of patiens. To aim at studying the effect of MDM2 and PTEN on the course of HCC carcinogenesis and development and its possible molecule mechanisms, which maybe provide the laboratory basis for the judge progrosis and clinical treatment of HCC.Methods The expression of MDM2 and PTEN was detected by SP immunohistochemical techniques in 60 cases of HCC, 27 cases of adjacent tissues and 10 cases normal liver tissues. to investigate relationships among MDM2 and PTEN and clinicopathologic factors of patiens. SPSS13.0 for windows software was used to analyse data. Difference of the ratio between groups was calculated byχ2 test or Fisher's exact test. Correlation was analysed by Spearman's rank correlation. The significant difference was considered when the P value was less than 0.05.Results (1) The positive protein expression of MDM2 was yellow or brown granule, MDM2 protein was mainly expressed in nucleus and partly in kytoplasm. The positive rate of MDM2 expression (31.67%) increased significantly in HCC tissues compared with that(7.41%) in adjacent tissues and that(0%) in normal liver tissues. The expression of MDM2 in HCC was related to portal embolus, capsule formation, number of tumor and TNM tumor stage (P<0.05), while not to sex, age, diameter of tumor, hepatocirrhosis, HbsAg, level of AFP and Endmanson grade (P>0.05).(2) The positive protein expression of PTEN was yellow or brown granule, PTEN protein was mainly expressed in kytoplasm and partly in nucleus. The positive rate of PTEN expression (56.67%) decreased significantly in HCC tissues compared with that(96.30%) in adjacent tissues and that(100%) in normal liver tissues. The expression of PTEN in HCC was related to tumor Endmanson grade, TNM tumor stage, portal embolus, capsule formation, number of tumor and level of AFP (P<0.05), while not to sex, age, hepatocirrhosis, HbsAg and diameter of tumor (P>0.05).(3) There were significant negative correlations between MDM2 and PTEN in HCC(rs=-0.423, P=0.001).Conclusions (1) The positive rate of MDM2 protein expression increased significantly in HCC tissues,The expression of MDM2 was related to carcinogenisis, invade and metastasize in HCC.(2)There is higher ratio of the low and lack protein expression of PTEN in HCC tissues , PTEN played an important suppressor role in the carcinogenisis, development, invade and metastasize of HCC.(3) Both of them promoted the carcinogenisis and development of HCC.(4) MDM2 and PTEN protein can be used as indicator to judge the biological behavior and prognosis of HCC.
