Objective:To establish the rat model of acute bacterial peritonitis and investigate theeffect of the intestinal pressure and the time of putting pressure on the intestinal endotoxintranslocation and the pathology of the rats, to observe the ways of the gut origin endotoxintranslocation from intestine to the body circulation ,the distribution of the gut origin endotoxinand the level of the serum endotoxin and the cytokine in abdominal infection rats, and toinvestigate the effect of QingReJieDu Herbs on the gut origin endotoxemia.Methods: We used the FITC-LPS as tracer and used the model of the rats intestine invitro to analysis and investigate the effect of the intestinal pressure and the time of puttingpressure on the intestinal endotoxin translocation and the pathology; At the same time, weused the FITC-LPS poured down the stomach as the tracer. The portal vein blood, the lymph inthe thoracic duct,the arterial blood, abdominal effusion, urine,stool samples and the tissuessamples of the liver, kidney, heart, lung, intestine, spleen were obtained to be determinedthe fluorescence intensity by the fluorescence and chemo-luminescence detection meter. Inaddition, the level of TNF—α,IL—6,IL—10,endotoxin, ALT, Cr, in serum and MPO of thelung tissue were detected, and investigate the effect of the QRJD on those indicators.Results: Higher pressure could damage intestinal barrier and promote intestinal endotoxintranslocation into blood circulation. Furthermore, the higher pressure could be promoted theLPS absorbed and seepaged. The level of the humor and the main organ tissue endotoxin ishigher than those of the control group. The order from high to low is lung, liver, kidney,intestine,heart, spleen. The indicators of the QRJD group were better than those of the modelgroup.Conclusion In the process of the intestinal endotoxin translocation, the higher gutpressure could damage intestinal mucosa barrier and putting pressure time had a coordinating action in the course of intestinal endotoxin translocation. Acute infectious peritonitis can lead to thetranslocation of intestinal endotoxin.. Gut origin endotoxin could be absorbed into the circulatorysystem via mesenterium lymph system, portal vein and peritoneum. The distribution of the gut originendotoxin in the main organ was different, especially in the lung, liver, kidney. QRJD herbscould significantly inhibite the translocation of gut origin endotoxin in peritonitised rats andQRJD herbs also could significantly decrease the level of serum endotoxin,IL-1,IL-6 and TNF-αin abdominal infection rats.
