| Syndrome complex of blood stasis is common symptoms in infectious abdominal diseases, Huoxue Huayu herbs has been believed as the main rule for the combined treatment of acute abdomen with traditional Chinese and western medicine. At present, however, there is no a Huoxue Huayu prescription which is based on treating infectious abdominal disease with accessible combination components, and sure clinical sure as well as clear mechanisms. In this study, we select four Huoxue medicines composed of Liangxue Huoxue Herbs which highly targeted for infectious abdominal disease. These four medicines are Sargentodoxa cuneata, Paconia Sufnuticosa, Paeonia lactiflora and Corydalis yanhusuo. Started with extraction of combination components, isolation and identification, the effect of Liangxue Huoxue Herbs is observed for acute bacterial peritonitis and abdominal adhesion formation. The corresponding mechanism was also studied.Methods:Using high-performance liquid chromatography (HPLC), Infrared Spectrometer and Mltraviolet Spectrometer, we quantitative analysized the compositions of the four Chinese herbal medicines, determined the combination components of the Liangxue Huoxue Herbs; animal model of acute bacterial peritonitis was established with cecal ligation plus puncture (CLP), and the histomorphologic change of organs after treated 3 days by Liangxue Huoxue decoction group and components group was observed. Moreover, blood samples were obtained for the detection of endotoxin and TNF-α,IL-1,IL-6. At the same time, we investigated the influence of Liangxue Huoxue herbs on intestinal permeability. Blood flow in intestinal mesenteric vascular, microcirculation and PO2 of intestinal tissues as well as rat model of peritoneal adhesions were also established with CLP, and the corresponding histomorphologic change of intestine treated 14 days by Liangxue Huoxue decoction group and components group were also observed. Moreover, blood samples were obtained for detection of endotoxin, fibrinogen and TNF-a, TGF-β1, ICAM-1, MMP-1, TIMP-1 and VEGF. At the same time, adhesive tissues were obtained for the detection of hydroxyproline(OHP), collagen and microvessel density(MVD). Results:The combination components of Liangxue Huoxue herbs include organic acid (chlorogenic acid), saponins (Sargentodoxa cuneata total saponin), terpenoids (paeoniflorin), essential oils (paeonol), polysaccharides (paeonol polysaccharide) and the alkaloid class (tetrahydropalmatine). The Liangxue Huoxue components were orepared from the Extraction, separation of the useful combination components. Treated with Liangxue Huoxue herbs for acute bacterial peritonitis, plasma endotoxin and serum cytokines were all decreased. It also can protect the intestinal barrier and increase blood flow in intestinal mesenteric vascular, microcirculation and PO2 of intestinal tissues in same degree. Treated with Liangxue Huoxue herbs for abdominal adhesion, protection of the intestinal barrier, plasma endotoxin and fibrinogen were all decreased. The contents of OHP and type I, III collagen were dramatically lower than others. At the same time, TNF-a, TGF-β1, ICAM-1, TIMP-1, VEGF and MVD were all decreased, and MMP-1 was increased. The same dose, the decoction group is better than component of group.Conclusion:the various combination components of Liangxue Huoxue herbs can treat acute bacterial peritonitis. The formation of abdominal adhesion are through many effective ways, including intervened blood circulation, the barrier function and the interaction of cytokines in the course of the formation of abdominal inflammation and adhesion, improved symptoms of acute bacterial peritonitis, and to reduce the formation of abdominal adhesions. Liangxue Huoxue herbs are an effective Prescription for treating infectious abdominal disease. |
