Objective To investigate the expression of epidermal growth factor(EGF)and epidermal growth factor receptor(EGFR)in the placental and fetal membrane in the rats with fetal growth restriction(FGR)and to cletect the effect and mechanism of ligustrazine ingert. Methods 24 pregnant rats were randomly divided into three groups:control,model,ligurstrazine(n=8 each).On day 20 Of gestation,FGR was induced by passive smoking and drinking in rats from the last two groups;ligustrazine was administered for the last group(Smg/Kg).The fetal rats were delivered by cesarean section.Birth weight,the lenth and poundera index of in the three groups were measured and compared.EGF concentrations in amniotic fluid and fetal blood from 24 pregnant rats were determined by radioimmunoassay(RIA).The expression of EGF and EGFR in placental and fetal membrance were analyzed by immunohistochemical assay.Results The body weight and height of FGR group were significantly lower than those in control group(p<0.05)in the FGR rats placenta and fetal membrance.expression of EGF were decreased(p<0.05).The expression of EGFR was increased(p<0.05).The treatment of ligustation injection the fetal growth and development in treatment group were close to that in normal control group.Conclusion 1.The model of FGR rats was made by means of passive smoking and drinking alcohol,which is a good animal model for FGR.2.The expression of EGF and EGFR were related to the FGR,Ligustrazin injection might improve the regulate the expression of EGF,EGFR and promoting fetal growth and development.3.Ligustrazine may impore placenta uterine volume of blood flow,prevention some causative factor for EGF and EGFR gene expression interception,promoting fetal growth and development.4.The pathological changes of FGR and the morphological base that lead to the hypofunction of placenta EGFR has important growth and development and FOR pathology process.
