Objective To analysis the sequence characteristics and the genetic polymorphism of the two new Y-STR Ioci(DYS715&DYS717)and obtain the allele and haplotype frequencies of them,.then discuss the possibility of applying them to forensic science by investigating 144 unrelated Han males of the two Y-STR loci in Shanxi population,to meet the need of more genetic markers in forensic practice and genetic analysisMethods The DNA were extracted by phenol/chloroform method,choosed two new Y-STR locui DYS715 & DYS717,and amplified with PCR,the products were analysised by agarose gel electrophoresis and nondenaturing polyacrylamide gel electrophoresis.Analysis the genetic polymorphism of 144 unrelated Han males in Shanxi population,23 female and 13 family were also carried out.Results The results show that the DYS715 locus consists of repeats of a core sequence(taga), five alleles were found,they are named 12,13,14,15,16 respectively with the allele frequency of 0.0833,0.4514,0.2292,0.1806,0.0556,and the gene diversity is 0.7060,the discriminating power and excluding probability of paternity are the same.The DYS717 locus contains two copies of a sequence motif,this motif has the following modular structure:(tgtat)2 tat(tgtat)(tgtac)m(tgtat)n,where the value of(m+n)ranges between 15 and 18 among different individuals,with the frequency of 0.0208,0.4444,0.3958,0.1389,the discriminating power is 0.6305.Among 144 Shanxi Han males,18 different haplotypes of these two loci were identified. Of these,4 occurred only once,with a frequency of 0.0069,and the most common haplotype occurred at a frequency of 0.2014.This system has a haplotype diversity(HD)of 0.8900 and the discriminating power(DP)of 0.8900.PCR products were not found in female DNA.The analysis of 13 father/son pairs has detected no mutation at the two locus.Different tissue from one corpse also has the same allele in the two new Y-STR loci,shows that the two Y-STR loci have a good tissue oneness.Conclusion These results indicate that the DYS715 and DYS717 loci are high genetic polymorphic,the haplotype diversity shows good discriminating power and high excluding chance of paternity.They are to be useful genetic markers in forensic science and human evolution studies.
