Objective To establish the rat models of diabetic nephropathy,and observe blood sugar, urinary protein/24h,renal function,pathological changes of renal tissue and exam blood plasminogen activator inhibitor-1(PAI-1)bioactivity in plasma in rats with diabetic nephropathy. To observe the effects of benazepril on PAI-1.The mechanism of diabetic nephropathy and renoprotective effects of benazepril on DN related to PAI-1 was specially studied.Methods 24 rats were randomly divided into control group(control group),diabetic nephropathy model group(model group)and group treated with benazepril(treatment group), every group had 8 rats.We established the rat models of diabetic nephropathy induced by streptozotocin(STZ).Three days after models'establishment,treatment was administrated (10mg/kg)by a stomach tube in treatment group,and same amount of water was fed in model and control group.Then we detected the blood sugar,urinary protein/24h,Scr,BUN,and used optics microscope to observe the the renal pathological changes.The expressions of PAI-1 and t-PA in plasma in rats.Results The blood sugar of model group rats which were injected STZ was higher than that of control group in 2w,4w,8w.Urinary protein/24h,Scr,BUN of model group rats were higher distinctly than those of control group(respectively,P<0.05).After treatment of benazepril,urinary protein/24h,Scr,BUN were reduced,(respectively,P<0.05).In the control group,they had low level of PAI-1 and high level of t-PA.In the model group,in contrast,the expression of PAI-1 in rats increased,and the expression of t-PA in plasma in rats was reduced(respectively,P<0.05).After treatment of benazepril,blood PAI-1 in rats was obviously reduced,and t-PA increased(respectively,P<0.05).Correlation analysis showed the expression of PAI-1 were positively relating to the damage of renal tissue.Their expression correlated with the renal injury(respectively,P<0.05).Conclusion Blood PAI-1 expression may increase in rats with diabetic nephropathy, which contributing to the occurrence and development of diabetic nephropathy.Benazepril have renoprotective effect on diabetic nephropathy by suppressing PAI-1 expression.
