| [Objective] To study the relationship between the mechanism of epilepsy and theregulatory genes of apoptosis by analyzing the expression of Bcl-2/Bax inhippocampus of rats after seizures.[Methods] 30 health Spague-Dawley(SD) male rats weighting 250 to 300g were usedand devided into control group, sham operated group, kainic acid(KA) injectiongroups (6h,24h,48h) at random. Each group has 6 rats. The epileptic model byintracerebroventricular injection of KA was used. The control group had none to dealwith. The sham operated group was used the same volume of artificial cerebrospinalfluid. The seizures were observed in the rats of each group. According to the criteriaof Racine, rats who met the criteria were chosed in the test. Rats of each group wereexecuted. HE staining and immunohistochemical staining were used. The change ofneuronal shape was observed and the number of the positive cells who expressed theprotein of Bcl-2 and Bax were observed and counted by the microscope. All theresults were analyzed by SPSS11.5.[Results]Rats by intracerebroventricular injection of KA can lead to seizures.Theseizures of the rats were coincidence with the criteria of Racine and the change of theseizures can be identified obviously.The results of HE staining show that the neuronalstain of the hippocampus in the control group, KA injection 6 hour group and thesham operated group is uniform and the form is normal. The neuronal stain of thehippocampus in the epileptic rats induced by KA injection groups (24h,48h) isabnormal that the volume of cell become small and the nuclear chromatin becomecondense, acidophilia stain is evidently. Compared with the control group and thesham operated group,the expression of Bcl-2 in KA injection groups(6h,24h,48h) increased(P<0.05), and the expression of Bcl-2 in KA injection 6 hour group is thehighest.The expression of Bcl-2 in KA injection 24 hour group and 48 hour groupdecreased. The expression of Bcl-2 in the control group and the sham operated grouphas no difference(P>0.05). Compared with the control group and the sham operatedgroup, the expression of Bax in KA injection groups(6h,24h,48h) increased(P<0.05),and the expression of Bax in KA injection 48 hour group is the highest. Theexpression of Bax in the control group and the sham operated group has nodifference(P>0.05). Compared with the control group and the sham operated group,theratio of Bcl-2/Bax in KA injection 6 hour group increased(P<0.05) and the ratio ofBcl-2/Bax in KA injection24hour group and 48 hour group decreased(P<0.05). Theratio of Bcl-2/Bax in the control group and the sham operated group has nodifference(P>0.05).[Conclusion] 1. The animal model by intracerebroventricular injection of KA is a goodepileptic model. 2. The phenomenon of apoptosis can be observed in the epilepsy.3.The Bcl-2/Bax poses influence on regulating and controlling over the apoptosis inthe epilepsy.In the earlier stage,the expression of Bcl-2 is higher than the expressionof Bax and neuronal structure of the hippocampus is normal. In the later stage, theexpression of Bax is higher than the expression of Bcl-2 and neuronal structure of thehippocampus shows apoptosis. The alteration of the Bcl-2/Bax ratio would play avital role in regulating the neuronal survival or death during epilepsy. |
