ObjectiveTo explore the protective effect of p38 mitogen activated protein kinase (MAPK) selective inhibitor SB202190 on hippocampal neurons in rats with kainic acid(KA)-induced intractable epilepsy.Methods60 SD rats were randomly divided into 6 groups, sham group,KA group, carbamazepine group and the SB202190 pretreated groups, inclouding the low dose group,the medium dose group and the high dose group. Behaviour was observed according to the criteria of Racine. Electrocorticogram was recorded by BL-420F.The number of apoptosis neurons was observed and the apoptosis rate was calculated by TUNEL.Expressions of p38MAPK,C-myc were detected by immunohistochemistry and Western blot.ResultsBehaviour observation according to Racine grading standard showed seizure-free in the blank group; the degree of attack was 5 in the KA group and it was 3~4 in CBZ group; while mostly 1~2 in the SB202190 prereated groups, Apoptosis cell reduced in the SB202190 treatment groups than those in the control group(P<0.01) and expressions of p38MAPK and C-myc were significantly decreased(P<0.01).ConclusionSB202190 has protective effect on neurons of KA-induced intractable epilepsy rats.The mechanism may be associated with expressions depression of p38MAPK and C-myc,respectively,in a dose-dependent manner.
