Objective: Amure is mainly distributed in the south area of Yunnan province. The root of the plant is used as a folk medicine for clearing away heat and toxic material, diuresis, treating of traumatic injury and arthritis. Its medicative for gout as a folk medicine has been watched in long clinical application. The studies on xanthine oxidase inhibiting activity of amure and its chemical constituents have not been reported. Studies on the chemical constituents of amure have been carried out for finding out the xanthine oxidase inhibiting constituents.Methods: Systematic solvents extraction has been made by Et2O, CHCl3, EtOAc, EtOH and water in succession. Column chromatography on silica gel and macro-reticular resine column have been carried out for the further separation and purification of those extracts of amure, 8 compounds have been re-crystallized. The structures of seven compounds of them were identified by classical chemical methods and spectroscopic methods analysis (MS, 1HNMR, 13 CNMR, DEPT). The structure of another one compound need being further identified. The xanthine oxidase inhibiting activities of these compounds has been investigated.Results: 8 compounds were isolated. The structures of seven compounds of them were determined as 2, 4-sorbic acid(â… ), Spinasterol (â…¡) , Daucosterol (â…¦), Taraxerone(â…§). Compoundâ…£,â…¤,â…¦showed xanthine oxidase (XO) inhibiting activities with IC50 value of 85.05μg/ml,97.50μg/ml,93.10μg/ml respectively in vitro.Conclusion: Eight compounds were isolated and separated from EtOH extracts of amure. Seven of thire chemical structures have been definited. Compoundâ…¥is a new natural product.
