Some of animals' kidney, liver, spleen, lung were collected for pathology examination to evaluate localized pathological change in a bodily organ or tissue.[Results]Amure ethanol extract reduced serum uric acid level after administration for 21 days in mice hyperuricemic models, the serum uric acid level showed significant difference between high dosage group and the model group(P<0.05) . However, serum uric acid level in high and low dosage groups both increased significantly(P<0.01) after administration for 14 days. Amure ethanol extract didn't show inhibitory activity on xanthine oxidase in liver. Amure water extract reduced the level of uric acid in plasma after administration for 14 days, and the descent of serum uric acid in high dosage group had statistical significance (P<0.05) , 21 days later, the serum uric acid of two groups both descended significantly (P<0.01) , and the UA average value of two groups reduced to normal level. Activity of xanthine oxidase in liver both decreased in two doses of Amure water extract administration groups(P<0.05) . The content of serum uric acid in Allopurinol-treated group decreased after administration for14 days and 21 days(P<0.05,P<0.01 respect-ively). Histological damages in treated group were improved in comparison with model group. Amure ethanol extract didn't increase the excretory amount of uric acid, and the excretion of uric acid in high dosage group even declined at 2nd and 3rd days after administration(P<0.05) . In the low dosage of Amure water extract administration group, the excretory amount of uric acid is more at 2nd day after administration than before administration(P<0.05) ,however, in high dosage group and benzbromarone-treated group, the excretory amount of uric acid is more at 1st day after administration (P<0.05) . Hyperuricemia model was induced successfully by feeding quails yeast and soybean flour. Serum uric acid level in managed group with yeast-soybean-Diclofenac was higher than control group (P<0.05) . Although there was no distinction between managed group with yeast-soybean and managed group with yeast-soybean-Diclofenac, the latter's serum uric acid level is lower. Two doses of Amure water extract, as well as Allopurinol, could all reduce the content of serum uric acid. Amure water extract had not obvious influence on glucose and triglycerides in plasma. There were not conspicuous histological changes among control group, model group and treatment groups.[Conclusions]Amure ethanol extract reduced the level of serum uric acid after long-term administration, however, it was formed independently of any proof that the effect was produced by inhibiting livers'xanthine oxidase or accelerating excretion of uric acid. The specific mechanism needed further approaching. Amure water extract reduced the level of serum uric acid significantly by means of not only inhibiting livers'xanthine oxidase but also accelerating excretion of uric acid. The histological damages of Amure-treated groups showed lesser degree than that of model group. The visceral damage resulted from yeast was not so severe as from adenine in hyperuricemia model. In short, Amure can be used to prevent and cure gout, hperuricemia and relative complications. The water extract is worthy of further being researched because of it's definite mechanism and significant effect on reducing serum uric acid level. |
