In our experiment, we try to explain how the liver maintain the liver functions as well as trigger the initiation of liver regeneration after loss of 2/3 of liver. Here differentially expressed proteins in rat livers at 1 h after partial hepatectomy (PHx) and sham operation were analyzed using two-dimensional electrophoresis combined with mass spectrometry. For each pooled sample in each group, 2-DE runs were repeated for reproducibility analysis. All spot comparisons and analysis were coordinated through the master image of PHx group and sham operation group, and the protein spots with two-fold changes or more in densities were considered to be different and were selected for further identification. 24 proteins identified by MS were classified into six groups according to their functional properties: a) lipid and energy metabolism; b) amino acid and purine metabolism; c) detoxifying enzymes; d) inflammatory factors and related proteins; e) axons regeneration; f) others. To confirm the fluctuation of the identified proteins, the differential expression of PDHX, PON1, Thyroid Hormone Receptor and GAP43 were validated by Western blot. Our data chiefly illustrated two aspects of remnant hepatocytes immediately after two-thirds volume hepatectomy. First, we found that blood sugar stabilization was maintained strivingly, main energy substrate was shifting from glucose to FFA, and the level of BCAAs may retainable in remnant hepatocytes, however, the synthesis of cholesterol and ketone body was decreased, nucleotide synthesis was learned mainly from the salvage pathway, the function of amino eliminating and biotransformation of residual liver did not recovered. Second, the relationship between host immunological response and the initiation of liver regeneration may be associated with each other, such as the increase of LBP and C3 could stimulate the trigger of regeneration, but the other up-regulated factors as IL2 suppressed it slightly, the result showed that the initiation of liver regeneration was complex but accurate, which may include more factors than all that have been documented.
