Object: To explore the molecular biological mechanism of YBKAL-capsule' anti-tumor effect.Methods: The model of S180 tumor-bearing mice was produced and treated with high, mid and low dosage YBKAL-capsule, and compound with CTX. After eleven days treatment, the experimental materials were drawn from mice and then mice were all sacrificed. Blood samples were obtained by extirpating mouse eyeballs. Tumor tissues were decoherenced and selected carefully, and were fixed by formaldehyde. Enzyme linked immunosorbant assay (ELISA) was employed to determine the amount of tumor necrosis factor a(TNF-α),interferonγ(IFN-γ) and interleukin 2(IL-2) in serum. The protein expression of Bcl-2 and P53 were evaluated by SABC immunohistochemistry.Results: (1) The effect of YBKAL-capsule to TNF-αin peripheral blood: High,middle and low dose of YBKAL-capsule groups were higher than the model group, middle and low dose of YBKAL-capsule groups compared with model group have significant difference(p<0.05). (2) The effect of YBKAL-capsule to IFN-γin peripheral blood. Compared with the model group, the content of IFN-γin different dose of YBKAL-capsule groups were significantly increased in peripheral blood, especially middle and low dose were higher than model group(p<0.05) . (3) The effect of YBKAL-capsule to EL-2 in peripheral blood: The different dose of YBKAL-capsule groups were all higher than model group, high and middle dose compared with model group have significant difference(p<0.01) , low dose was higher than model group(p<0.05). (4) The effect of YBKAL-capsule to the expression of bcl-2,p53. The protein expression of p53 and bcl-2 in each group was less than model the group.Conclusion: YBKAL-capsule can increase the content of TNF-α, IFN-γand EL-2 in peripheral blood; it can also inhibit the gene expression of p53 and bcl-2. The anti-tumor mechanism of YBKAL-capsule maybe concerned with enhancing the function of immune system and inhibiting the gene expression of p53, bcl-2.
