Objective Detect the polymorphism in 5A/6A MMP-3 gene promoter in 104 Han Lanzhou patients with essential hypertension and 102 healthy adults through polymerase chain reaction (PCR) - restriction fragment length polymorphism (RFLP), to investigate the possible impact of the 5A/ 6A polymorphism on the risk of essential hypertension in Han Lanzhou area population.Methods The study enrolled 104 hypertensive patients and 102 control subjects without hypertension excepting other relative diseases. The diagnosis of hypertension were confirmed according to World Health Organization criteria. MMP-3 promoter gene containing the 5A /6A polymorphism was amplified by polymerase chain reaction (PCR). PCR products were digested by PsyI and then were separated by electrophoresis on agarose gel.Results The distribution of MMP-3 genotype was not significantly different between hypertensive patients and controls. The genotype frequencies among hypertensives were 5A/5A 0.01, 5A/6A 0.336 and 6A/6A 0.654, the allele frequencies of 5A 0.178, 6A 0.822. In controls, 5A/5A0.02, 5A/6A 0.441 and 6A/6A 0.539, 5A 0.240, 6A 0.760. After dividing Study sample by gender, distribution of MMP-3 genotype and allele frequencies also were not significantly different between hypertensive and normotensive neither males nor females( P<0.05). When Multiple logistic regression analysis was used, we found 5.97-fold increased risk for hypertension associated with genotype 6A/6A (P =0.013) in young males (<50 years) according to recessive model.Conclusion These results indicate that there was gender and age-specific differences in association of MMP-3 5A/6A polymorphism with hypertension in Lanzhou Han population. The 6A allele is likely to be important factor in the development of hypertension in young males.
