Acne can be say comedo, zit, is a chronic inflammatory disease of the pilosebaceous follicles, caused by many factors and characterized by comedones, papules, pustules, cysts, nodules, and often scars. It is a disease of the adolescent, with more than 90% of all teenagers being affected to some degree. In moderately severe cases, disfigurations occur. 15% of patient's condition is more serious and required therapy. Though there are many therapy ways, so far there is no an ideal way that all of people admittedly.Chemical peeling is an in-office procedure that involves the application of a chemical agent to the skin to induce controlled destruction or exfoliation of old skin and stimulation of new epidermal growth with more evenly distributed melanin. When peel agents reach the dermal layer, important wound-healing activities occur that cause skin remodeling and skin smoothing, both antiaging benefits. Classification of peel depths: ①Superficial peels, destruction anywhere from the epidermis to the papillary dermis; ②Medium peels, destruction extending to the upper reticular dermis; ③Deep peels, destruction extending to the mid-reticular dermis.Alpha hydroxyl acids (AHAs) are a group of organic acids that have recently become popular in the treatment of a variety of skin conditions, particularly those characterized by hyperkeratinization. The exact mechanism of action of AHAs is not completely understood, it has been demonstrated that these acids can gently break the bonds between cells in the stratum corneum which enables exfoliation, remove the uppermost skin layers which enhances the natural process of exuviations, disperse basal layer melanin, and increase collagen synthesis within the dermis. Now more and more doctors use AHAs peels for treatment of acneand it works well. We design the experiment of the in vitro bacteriostasis effects of Propionibacterium acnes by AHAs and establish an experimental animal model for acne to observe the anticomedone effect of AHAs for more know how AHAs work.Objective: Research and validate the work of AHAs treating acne and make it apply far and wide by testing the effect of the in vitro bacteriostasis effects of Propionibacterium acnes by AHAs and observing the anticomedone effect of AHAs.Methods: Observed the Minimum inhibitory concentrations (MICs) of AHAs to Propionibacterium acnes by small volume dilution method with the National Committee for Clinical Laboratory Standards (NCCLS) approved M11-A5—Methods for Antimicrobial Susceptibility Testing of Anaerobic Bacteria. According to Kligman's method , coal tar was applied daily to the area nearby the opening of albino rabbit ear canal for two weeks .Then ,some rabbits was treated with AHAs, the others not. At last we observe the anticomedone effect of AHAs.Result: AHAs has better sensitivity to Propionibacterium acnes and the MICs of AHAs to Propionibacterium acnes is 0.73-1.46mg/ml. Acniform lesions on the rabbits' ears could be seen two weeks later, which were similar to the findings of acne in human being and under optic microscope. The ears acniform lesions which had been cured improved, the others no change.Conclusion: 1. AHAs can well inhibit Propionibacterium acnes in vitro, which is the main pathogenic bacteria that lead to acne. 2. This animal model of acne can be repeated and might be used to observe the effects of drugs. 3. AHAs could reduce the follicular keratinous plug and the dermal inflammation. AHAs can inhibit both folliculus pili dyskeratosis and the growth of Propionibacterium acnes, it acts on the two main facts which lead to acne so AHAs is a valid method to treat and prevent from acne.
