Veronica sibirica (L.) Pennell (Scrophulariaceae) is one folk medicinal plant which is occuring mainly in most northern parts of China. It has been used as a medicinal herb to cure influenza, throat pain, parotitis and rheumatism in clinical. It has the pharmacological effects of dephlogisticate and odynolysis, and many chemical constituents were reported including ferulic acid, 3,4-dimethoxy cinnamic acid, 3-O-acetyloleanolic acid, D-mannitol, dancosterol, etc. Other relative researches have not been taken so far.In order to search the bioactive constituents from nature source, we began to study the chemical constituents from V. sibirica. By column chromatography, including Silica gel, Toyopearl HW-40, Sephadex LH-20 and preparative HPLC, three new (1-3) and five known compounds (4~8) were isolated. By means of 1H-NMR , 13C-NMR and 2D-NMR spectra, their structures were identified as sibiriquinone A (1), sibiriquinone B (2), 1,2-dehydrocryptotanshinone (3), ferruginol (4), dihydrotanshinoneⅠ(5), tanshinoneⅠ(6), tanshinoneⅡA (7), and cryptotanshinone (8), respectively.The P.E, EtOAc, BuOH extracts and isolated compounds, were screened by antitumor activity experiments. The P.E, EtOAc and BuOH extracts all have strong cytotoxicity against TJ-905,Hela,SK-N-SH and BEL-7402 cells. Compounds 1, 2, 4~8 revealed specific antitumor effects on different cancer cells. Compounds 5 and 8 show dose-depending bioactivities, and the IC50 of 5 on TJ-905 and Hela was 1.038μg/ml and 1.816μg/ml, the IC50 of 8 on TJ-905 and Hela was 3.637μg/ml and 4.391μg/ml, respectively. The flow cytometry analysis showed the P.E extract had no influence on Hela's cell cycle, and compounds 5, 7 and 8 blocked Hela cells in S phase.In the lymphocyte transformation experiment, the valve of inhibition percent of the P.E, EtOAc, BuOH extracts and compounds 1, 2, 4~8 revealed a significant activities in comparison to the concanavalin A control group (p<0.01).
