Study On Effects Of Reduction Fat Deposition In Experimental Obesity Rats By Antibodies Against Adipocyte Membrane Proteins

In order to verify the effect of polyclonal antibodies against adipocyte plasma membrane proteins on reducing fat deposition,antibodies were firstly prepared in rat against purified plasma membrane proteins from porcine adipocytes and the pattern of fat SD rats was founded in this experiment,which would afford theory evidence and experiment data.This experiment chosed 75 SD rats,weighting approximately 200g.75 SD rats were allocated randomly to five groups(fifteen rats in each).GroupⅠwas used as control group for founding the fat pattern and fed with lower energy feed,groupⅡ,Ⅲ,Ⅳ,Ⅴwere used as experiment group and fed with higher energy feed.After the fat pattern had founded,GroupⅡwas used as control group for antibodies injection experiment and intraperitonealy injected with physiological brine, groupⅢ,Ⅳ,Ⅴwere used as experiment groups and injected with 0.1,0.5 and 1.0 mg antibodies,respectively.The result indicated that:Polyclonal antibodies against adipocyte plasma membrane proteins can increase growth performance of fat rats.After 6 weeks feeding,compared with control group(groupⅠ),added weight of experiment groups fed with higher energy increased 26.91%,26.72%,20.85%and 21.08%(p<0.05)and reached advisable fat degree;the content of fat metabolism index CHO,HDL,LDL were lower and reached remarkable difference,the content of TG decreased 26.40%(p<0.05),4.80%,9.60%(p>0.05)and 18.40%(p<0.05);the content of endocrine metabolism index Ins was higher than that of groupⅠ(p<0.05),IGF-I,GH,T3,T4 were lower (p<0.05).After injecting fat patten rats with different quality antibodies,the content of fat metabolism index were higher than that of control group.The content of CHO,TG,HDL and LDL increased at different extent from the first week after immunized with antibodies and maintained to over the experiment.At the end of the experiment,compared with control group, the CHO content of groupⅢ,Ⅳ,Ⅴincreased 29.46%,61.61%and 93.75%(p<0.05),respectively;TG increased 37.89%,31.58%(p<0.05)and 14.74%(p>0.05);HDL of groupⅣ,Ⅳ,Ⅴincreased 91.11%and 46.67%(p<0.05),LDL increased 75.00%,68.75%and 56.25%(p<0.05),respectively.The Ins content of experiment groups began to drop after injecting with antibodies and maintained to the end of the experiment,compared with control group,Ins decreased 45.14%,47.22%and 43.75%(p<0.05);The content of triiodothyronine(T3)and tetraiodothyronine(T4) increased,T3 increased 50.00%,39.11%and 37.27%(p<0.05),T4 increased 2.27%,18.18% (p>0.05)and 27.27%(p<0.05);IGF-I were higher than that of control group and increased 13.12%,5.57%and 6.21%(p>0.05)at the end of the experiment,respectively;GH was higher than that of control group at every half an hour in next day after immunized with antibodies and increased 27.27%,90.91%and 27.27%(p<0.05),GH were higher than that of control group at experimental period and maintained to the end of the experiment,the content of GH increased 76.92%,61.54%and 92.31%(p<0.05)at the end of the experiment,respectively.Polyclonal antibodies against adipocyte plasma membrane proteins can significantly improve carcass composition of fat rats.Compared with control group,carcass weight of experiment groups had not significant difference;fat percentage decreased 13.62%(p>0.05),28.14%and 37.78%(p<0.01);Lean meat rate increased 19.25%(p>0.05),27.21%and 23.02%(p<0.05);The content of attach-spermary fat decreased 13.75%,16.33%and 20.34% (p<0.05),respectively;hypodermic fat of groupⅣ,Ⅴdecreased 20.27%and 35.41%(p<0.05), groupⅢhad not significant difference;Intraperitoneal fat decreased 21.49%,47.79%and 58.20%(p<0.05),respectively.
