Background : At the present, experiment has proved that the activity of Acanthopanax Senticosns Saponins have protective effects on myocardial ischemia. But ,It is not clear that Ciwujianoside-B(Sb) has protective effects on myocardial ischemia.The research about this still have not been reported.Aim: We wil research that ciwujianoside-B protect against acute myocardial ischemia of rats caused by isoproterenol.Methods: The first,the methods of leadⅡECG was used to observe effect of Sb on anesthetied rat and acute myocardial ischemia model on rat. Then, The methods of myocardial mechanics on heart was used to observe effect of Sb on anesthetied rat and acute myocardial ischemia model on rat.The parameters of myocardial mechanics including left peak-value systolic pressure (LVSP), left ventricular end diastolic pressure (LVEDP), beat rate (HR) ,maximum ascending and declining rate of left ventricular pressure (±dp/dtmax) were recorded with 8-channel polygraph at same time .Finally,in order to exclude the impact of nerves and body fluids, the methods of Langendorff was used to observe effect of Sb on anesthetied rat and acute myocardial ischemia model on rat.Results :1. Firstly,intraperitoneal injection of isoproterenol after 10 min, S-T segment of Rats promoted from 0.026+0.006mv to 0.188+0.023mv.The increase was more than 0.1mv , as evidenced by the successful model of myocardial ischemia. After injection Sb(10mg/kg) in tail vein of myocardial ischemia rat ,the increase of S-T segment was inhibited,it was dropped to 0.097+0.026mv. 2.Secondly, high concentrations of Sb reduced significantly in rats LVSP and±dp/dtmax, lower rate of 20.81%,17.12% and 30.19%, but no significant effect on HR and LVEDP. Intraperitoneal injection of isoproterenol after 10 min, LVEDP and HR of rats increased significantly,increasing rate of 14.69% and 220.51% .LVSP and±dp/dtmax decreased significantly, lower rate of 21.92%.19.75% and 48.54%,they reached significant levels than control. After injection Sb(10mg/kg) in tail vein of myocardial ischemia rat , changes of myocardial mechanics coused by isoproterenol were significantly weakened. Increasing rate of HR and LVEDP were 6.47% and 71.79% respectively,compared with the model group significantly lowered. Lower rate of LVSP and±dp/dtmax were 10.07% ,1.36% and 26.17%, the variations were significantly lower than model group.3.Sb (500 mg/L) can reduce LVSP and±dp/dtmax of isolated rat heart ,lower rates were respectively 22.56%,37.99% and 53.2%. LVEDP was increased. The increasing rate was 33.33%.When after 10 min reperfusion isoproterenol (50mg/L),LVEDP and HR of isolated rat heart...
