Uterine leiomyoma,which is also called myoma of uteires, is the most common benign tumor for women at reproductive ages. But its conecrete etiology is not defined. It has been approved that uterine leiomyma is the desease that depending on sex hormone by earlier researching, and estrogen and progesterone play an important role in the growth of the disease. Recent reseach results indicate that the occurrence of uterine leiomyomas is closely related to the cell proliferation and abnormal apoptosisCell apoptosis is the main form of cell death, which plays an integral role in the development and maintenance of multicellular organisms. In normal physical condition, there exist a balance between cell apoptosis and cell proliferation, Once the balance is destroyed, it may lead to many diseases. Cell apptosis has the effects in embryoonic development, stable of internalenvironment, immuno logic-drangement, neuropaty, auto-immue disease and occurrence of tumor. In many research on the relationship of apoptosis gene and uterine leiomyoma, the bax gene research has been paid more attention to. The bax gene, namely bcl-2,is connected by x protein gene ((bcl-2-associated)x protein gene).It can resist the bcl-2activeness, which has the function of promoting cell proliferation. Once the cell proliferation happens, the nucleic protein and DNA interactination is breakdown,which causes to DNA inapopotosis cell beginning to be sensitive.As a result, denaturation, for the enough length, forming certain conformation single chain DNA, may combine with the anti-ssDNA immune body.In this experiment ,Immuno-histochemical S-P staining was used to test single-strand DNA and the protein expression of bax gene in both uterine leiomyomas and normal myometria adjacent to leiomyomas of 40 cases with uterine leiomyoma by anti single-strand DNA (ssDNA) antibody and anti Bax monoantibody to approach the association of the uterine leiomyoma genesis with cell apoptosis as the significance detecting of the cell apoptosis in uterine leiomyoma by anti single-strand DNA (ssDNA)antibody.Results:bax (p-19) is maily located in endochylema presented as yellow partidese, In 40 cases SMCs, 38 cases have bax (p-19) presentation. In total, 84.2% are mid range to strong expression. In UL, The positive rates and intensity of immunostaining Bax are much more lower in the uterine leiomyomas than those in the normal myometria ( P<0.01).The reasen is that chain DNA that is destroyed to be the cell DNA forms exist in the apoptosis cell. In 37 normal myometrium samples, the SMCs nucleus aim directly to ssDNA antibody to present as yellow partidese; The ssDNA masculine sample ratio reaches 92.5%(37/40), remarkable is higher than 62.5%(25/40)in uterine leiomyomas (X2=10.323.p=0.001), in which 84.2% (32/38) is strong masculine expression; Minority is weak expression. But mainly expresses in um for the weak masculine gender (64%,16/25). Mediumobviously is lower than normal myometria smooth muscle sample in fibroid sample ssDNA to the strong masculine expression rate (X2 =14.799, P< 0.01). And, ssDNA assumes the weak expression in the SMCs the accent to small apoptosis body mainly to concentrate in on the womb smooth muscle myo- micrangium wall, but in the UL,it has not seen the similar situation occurs.The positive rates and intensity of immunostaining both single-strand DNA and Bax are much more higher in the normal myometria than those in the uterine leiomyomas ( P<0.01). Moreover, there is a positive correlation between the quantity of Bax protein expression and the amount of the single-strand DNA (r=0.720,p =0.000).Conclusions: Down-regulation of Bax expression may play an important role in genesis of the uterine leiomyoma, therefore the absence of cell apoptosis in uterin smooth muscle should be a t reason of inducing uterine leiomyoma. Apoptosis cells in uterine smooth muscle could be detected exactly by anti-ssDNA antibody.
