Purpose:With the application of enzyme technology and Image Analysis,we study the gastrocnemius of the mouse,the changes of the endplates,and the impact on the muscle fibers and the neuromuscular junctions of the swimming mice.Methods:As experimental objects, 30 Kunming male mice that each one weight 34±1.87g with feeding by the state standards of rodents feeding and free eating influent are divided into 3 groups randomly after a week feeding adaptabily, that is, the comparison group, the unload group, and the load group.A. The comparison group is immersed in the same temperature shallow water and limited them's activities. B. the unload group swim by medium intensity for training. The swimming pool size is 150 cm x 32cm x 80cm and 25 cm depth. The temperature of water is 32±1°.C. They are trained six days a week and trained once in a day. The first day trained for 10 minutes and since then gradually increased to 10 minutes the first time to swim at the weekend reached 60 minutes. Second weekend to 120 minutes. Intensity beyond this swim for eight weeks. C. the load group is trained for 120 minutes with the group training conditions and methods of the same situation. Only the tail load increased 5% of the weight (fine wire winding tail). After swimming training eight weeks, double hindlimb muscle biopsy done frozen quickly with 4% intraperitoneal pentobarbital anesthesia. Acetylcholinesterase (AchE). succinate dehydrogenase (SDH), adenosine triphosphatase (ATPase) were analysised with enzyme technique and using image analysis system.Results:After eight weeks of swimming training, it showed that compared with the control group,the percentage of ST in the load groups rose significantly.A significant difference(P<0.05) was displayed which the red muscle fibers in the percentage of the area of the the load group and the control group. On the contrary,the percentage of FT in the load groups reduced.A significant difference (P<0.05) was displayed which the load group compared with the control group decreased significantly. The red muscle fiber diameter than the control group of the load group increased significantly. Compared to the red muscle fiber diameter of the load group and the control group, it has a very significant difference (P<0.01). White muscle fiber diameter reduced which exercise group compared with the control group decreased significantly, a significant difference was displayed. The motor endplate diameter of the three groups had increased trend. Control group compared to the the load group showed a significant difference (P>0.05). The image analysis statistics showed that the SDH optical density of the the load group was significantly increased. A significant difference was displayed compared with the control group (P<0.01). Average integral optical density of the unload group was significantly increased.A significant difference was displayed (P<0.05). The ATPase average integral optical density of the load group was significantly lower. A significant difference was displayed compared with the control group (P<0.01). The ATPase average integral optical density value of the unload group lowered. There was statistics significant that a significant difference was displayed (P<0.05). Conclusion:Swimming training with loads promotes transformation of the muscle fiber types and increases the diameters of the muscle fiber and the endplate as well.
