Objective To study the effect of iodine on skin wound healing in rats, the expression of VEGF and bFGF in the wound tissue and its ultrastructural changes. Methods A total of 40 Wistar rats were selected. Bilateral round wounds were made on the backs of the rats by corneal trephine and each rat was made six wounds. The six wounds of each rat were divided randomly into six groups, and there are bFGF-treated group, Sodium Chloride-treated group, NI-treated group, 10HI-treated group , 100HI-treated group, 1000HI-treated group. The effect of Iodine on the skin wound was evaluated through determining closure time and contracting ability of skin wound, detecting the expression of VEGF and bFGF in the wound tissue by immunohistochemical techniques and image analysis system and observing histopathological characteristics and its ultrastructural changes. Results The results show that there was significant difference about healing rate among six groups. 10HI-treated group was significant higher than Sodium Chloride-treated group (P<0.05 or P<0.01 ) and there was no obvious difference between 10HI-treated group and bFGF-treated group. Light microscope showed that in the 10HI-treated group and bFGF-treated group, there were more neoformative blood vessels on the 7th day and there was no obvious distinction between these two groups. In addition, when compared with the Sodium Chloride-treated group, VEGF, bFGF of bFGF-treated group and each using Iodine group increased by immunohistochemistry(P<0.05 or P<0.01), together with ultrastructural organization changes were significant in the 10HI-treated group and bFGF-treated group. Conclusion We find that: â‘  Befitting iodine had the ability of promoting skin wound healing in rats, and the higher concentration of iodine could damage it. â‘¡ There were more neoformative blood vessels in the 10HI-treated group and bFGF-treated . â‘¢ Befitting iodine could promote the expression of VEGF and bFGF in the wound tissue accordingly, promote wound healing.
