| Purpose: Attempted isolation of nanobacteria from upper urinary tract stones and detected by means of microscopy, to determine whether the nanobacteria exists in upper urinary tract stones.Materials and methods: 17 upper urinary tract stones were remove from ten patients who had undergone pyelolithotomy or nephrolithotomy or urterolithotomy or ureteroscopy. The stones were processed and subjected to mammalian cell culture. After 4-5 weeks, the culture were examined using a phase contrast microscopy, transmission electron microscope (TEM) , Scanning electron microscope(SEM). Hoechst33258 stained the DNA of the nanobacteria and examined using fluoroscopy. Results: after 3 weeks, 17 stones could see nanobacteria ,and more obviously after 4 weeks by means of a phase contrast microscopy .the cultures after 4-5 weeks could observed the nanobacteria using transmission electron microscope ( TEM ) , Scanning electron microscope(SEM). After staining, fluoroscope did not observed the obviously presence of DNA. Conclusions: the nanobacteria exists in upper urinary tract stones andmaybe play a role in the formation of stone. |
