BACKGROUND&OBJECTIVE: Estrogen directly up-regulates LRP16 gene expresson via activating its receptor (ER) , and the overexpression of LRP16 promotes the proliferation of human breast cancer cells.Liver is also a hormone-depended-on organ.This study was to detect the protein level of LRP16 gene in hepatocellular cancer, and investigate its correlation to the clinicopathologic features.METHODS: The protein level of LRP16,ER in carcinoma,matched peritumor tissues and normal hepatocellular tissues from 42 hepatocellular carcinoma patients were detected by Western blot.The expression of CD34, P53, VEGF, in the carcinoma tissues were detected by immunohisto chemistry.RESULTS: According to the results of Western blot, compared with that in peritumor and normal tissues, LRP16 was lower-expressed in carcinoma tissues.Of the 17 samples with LRP16 higher-xpression , 12 were ER-positive, 5 were VEGF-positive,4were P53-positiveand5 were MVD-positive; of the 25 samples without LRP16 overexpression, 3 were ER-positive , 17 were VEGF -positive, 14were P53-positiveand21 were MVD-positive.The positive rates of LRP16 were significantly higher in the samples without LRP16 overexpression than in the samples with LRP16 overexpres sion (P<0.05 ) Significant associations were found between LRP16 and the number of nodules, tumor size, Edmonson-Steiner class and ect. CONCLUSION: LRP16 was lowered-expressed in carcinoma tissues, compared with that in peritumor and normal tissues. LRP16 overexpression is closely correlated to the positive rates of ER and lessermalignantbiological behavior.Both ER and LRP16 status are good prognostic factors.
