Objective: EGFR is an important receptor of modulating epidermal cell differitation; Bcl-2 is the most important apoptosis inhibit factor, Fas is an important apoptosis enhance factor, both of them have a close relationship of cell apoptosis.To invest the role of EGFR, Bcl-2 and Fas in some proliferative cutaneous lesions , We detected the expression of those factors in normal epidermis, lichen planus(LP), seborrheic keratosis(SK) and basal cell epithelioma(BCC) .Methods: Thirty-four paraffin embedded lesions are collected for diagnostic histopathology from the files of Dermatologic clinic , the second affiliated hospital of DaLian Medical University. Specimens include 10 LP ,12 SK, 10 BCC. 10 normal skin from plastic surgery were used as control. The distribution and intensity of EGFR,Bcl-2,Fas of 44 specimens were detected by immunohistochemical SP method.Results:â‘ Normal Skin: EGFR was expressed in basal layer and part of spinous layer ; Bcl-2 was positive only in basal layer; Fas was expressed in basal layer, spinous layer, sebaceous glands and outer root-sheath.â‘¡LP: EGFR was expressed in basal layer, spinous layer and stratum granulosum and had an obvious increase than normal skin; Bcl-2 was positive in basal layer; spinous layer,sebaceous glands and had a higher expression than normal skin;Fas was positive Significant increase than normal skin and had an diffusion expression in the whole epidermal.â‘¢EGFR was expressed generally in upper level of spinous layer ,stratum granulosum and stratum corneum and was obvious higher than normal skin; Bcl-2 was positive in basal layer; Fas was positive in basal layer and sebaceous glands.Both of them had no different expression than normal skin.â‘£BCC: EGFR was expressed diffusionly in tumor cell clumping and had an higher expression in periphery tissues than central one; Bcl-2 was positive diffusionly in tumor cell clumping; both of them had an significant increase than nomal skin. Fas positive staining protein was diffused distribution in tumor cellclumping.There is no difference between BCC and normal skin.Conclusions:â‘ EGFR, Bcl-2, Fas were expressed in patients with LP and had a significant increase than normal skin. This result suggest that EGFR hyper-expression in LP may be related to cell proliferation and apoptosis inhibition and promotion exsisted at the same time .â‘¡EGFR was expressed in SK and had a significant increase than norma skin,while Bcl-2 and Fas were expressed possibly slightly.This result suggested that cell proliferation in SK may be related to EGFR hyper-expression and may have less relationship with the apoptosis inhibit function of Bcl-2 .â‘¢The expression of EGFR and Bcl-2 in patients with BCC was significantly higher than in normal skin while expression of Fas was weak. This result suggested that tumorigenic proliferation of BCC may be related to EGFR hyper-expression , enhancement of the effect of apoptosis inhibition of Bcl-2 and decrease of the effect of apoptosis enhancement of Fas.
