Objective To study the effect of vitrification solutions with cryoprotectants EG singly or DMSO together for the cryopreservation of mouse blastocysts . To compare the difference of vitrification solutions adding Ficoll 70 or not. To find out the change of ultrastructure for cryodamage of expanded blastocysts.Methed 1. To promote 6-8W small mouse ovulate and obtain early blastocysts and expanded blastocysts.2. To cryopreserve early blastocysts and expanded blastocysts respectively with six kinds of vitrification solutions (ES35, EDS35, ESF10, ESF20, EDSF10 and EDSF20) . 3 To observe the ultrastructure of frozen-thawed and fresh expanded blastocysts at electron microscopic level.Results 1. As to the survival and hatching rate of early blastocysts , EDSF10 group is the highest among the six groups, and there is no significant different with the control group, but ES35 group is the lowest and is statistical different with EDSF10 group and the control group. 2. As to the survival rate, there is no significant difference between every experimental groups , but there is significant difference between experimental groups and the fresh group. The hatching rate of expanded blastocysts of EDSF10 group is highest and ES35 group is the lowest among the six groups, but they are both statistical different with the fresh group. 3. EG^DMSO is statistically helpful to the survival and hatching rate of early and hatching blastocysts. 4. Ficoll 70 is statistically helpful to the survival rate of early blastocysts, and 10% is enough for vitrification. 5.Except for EG35 group,the survival rate of early blastocysts are higher than expanded blastocysts;for EDS35, ESF20 and...
